In 1941, bugler and career soldier Private Robert E. Lee Prewitt (Montgomery Clift) transfers to a rifle company at Schofield Barracks on the island of Oahu. Peter Parker plans to propose to Mary Jane Watson, who has just made her Broadway musical debut. A meteorite crashes near the two in Central Park, and an extraterrestrial symbiote attaches itself to Peter's moped. While fleeing police, escaped prisoner Flint Marko falls into a particle accelerator that fuses his DNA with surrounding sand, allowing him to shapeshift at will as the Sandman. Peter's best friend, Harry Osborn, who blames him for his father's death, attacks Peter using weapons based on his father's Green Goblin technology. Harry injures his head and suffers partial amnesia, forgetting that Peter is Spider-Man and his desire for revenge. During a festival honouring Spider-Man, Marko robs an armored car. NYPD Captain George Stacy tells Peter and Aunt May that Marko was Uncle Ben's killer, and Dennis Carradine was only an accomplice. While a vengeful Peter sleeps, the symbiote bonds with him. Peter wakes up hanging from a skyscraper. His costume becomes black and his powers increase, but the symbiote brings out Peter's dark side. Wearing the new suit, Spider-Man locates Marko and battles him in a subway tunnel, reducing him to mud using water.

High-school senior Peter Parker visits a genetics laboratory with his friend Harry Osborn and love interest, Mary Jane Watson for a school field trip. "Bernie" LaPlante is a pickpocket and petty criminal who anonymously rescues survivors of an aircraft crash, in order to steal some of their purses and wallets – losing a shoe in the process. He later meets a homeless Vietnam veteran, John Bubber, tells him about the rescue at the crash site and gives him his remaining shoe. When a television station offers $1 million to the "Angel of Flight 104", Bernie cannot claim the reward, due to his arrest while fencing credit cards he stole from the people he rescued. Bubber, however, uses the single shoe to take credit for the act. Bernie tries to tell people that Bubber is a fake, but the media has sensationalized the heroic Bubber, and everyone wants to believe in him rather than Bernie.

Bob Wiley (Murray) is a good-natured man with great work ethic, but he suffers from multiple phobias and is divorced because his ex-wife is a fan of Neil Diamond. He feels good about the results of an initial session with Dr. Leo Marvin (Dreyfuss), a New York psychiatrist with a huge ego, but is immediately left on his own with a copy of Leo's new book, Baby Steps, when the doctor goes on vacation to Lake Winnipesaukee, New Hampshire for a month. Unable to cope, Bob follows Leo to his vacation home. Leo is annoyed because he does not see patients on vacation but, seeing how desperate Bob is, he gives Bob a prescription telling him to "take a vacation from his problems." Dominick "Nicky" (Hulce) and Eugene "Gino" (Liotta) Luciano are twin brothers living together in Pittsburgh, Pennsylvania.
