A girl named Riley is born in Minnesota, and within her mind, five personifications of her core emotions—Joy, Sadness, Disgust, Fear, and Anger—come to life. The emotions live in Headquarters, Riley's conscious mind, where they influence Riley's actions and memories via a control console. Her new memories are housed in colored orbs, which are sent into storage at the end of every waking period. The most important or "core" memories are housed in a hub in Headquarters and power five "islands", each of which reflects a different aspect of Riley's personality. Joy acts as the dominant emotion to keep Riley in a happy state, but she and the others do not understand Sadness's purpose, other than making Riley cry.

High-school senior Peter Parker visits a genetics laboratory with his friend Harry Osborn and love interest, Mary Jane Watson for a school field trip. However, it also drives him insane and he kills his assistant, Mendel Stromm. The next morning, Peter finds that his previously impaired vision has improved and his body has metamorphosized into a more muscular physique. At school, he finds that his body can produce webs and his quickened reflexes let him avoid injury during a confrontation with bully Flash Thompson. Later, Fogell (Christopher Charles Mintz-Plasse) comes up to the two and reveals his plans to obtain a fake ID during lunch. Seth uses this to his advantage and promises to bring alcohol to Jules' party. Fogell's fake ID is met with derision by Seth and Evan, as it states that Fogell's name is simply "McLovin". After contemplating their options, Seth decides they have no choice but to have Fogell buy the alcohol with his fake ID. Fogell goes in and successfully buys the alcohol, but is interrupted when a robber enters the store, punches him in the face, and takes money from the cash register.

A bomb detonates in the plaza of the Colombian Consulate building in Los Angeles, killing nine people, including a caravan of Colombian officials and American intelligence agents. Among the civilians killed are the wife and son of LAFD firefighter, Captain Gordon "Gordy" Brewer, who was injured in the explosion. A tape is sent to the U.S. State Department, in which a masked man calling himself "El Lobo" (The Wolf) claims responsibility, explaining it was in retaliation for the oppression of Colombia by the United States. The FBI believes El Lobo is a Colombian terrorist named Claudio Perrini. CIA Special Agent Peter Brandt, the Colombia Station Chief, is harshly reprimanded for the incident by a Senate Oversight Committee, who promptly terminate all CIA operations in Colombia. Brandt angrily returns to Mompós and meets with his paramilitary allies to plan a major offensive to take down Claudio.
