A dog named Bolt (John Travolta) and a girl named Penny (Miley Cyrus) star on a hit television series called Bolt in which the titular character has various superpowers and must constantly thwart the evil plans of the nefarious Doctor Calico (Malcolm McDowell). To gain a more realistic performance, the TV show's producers have deceived Bolt his entire life, arranging the filming in such a way that Bolt believes the television show is real and he really has superpowers, including a powerful sonic scream-like "superbark". Mistakenly believing Penny has been kidnapped by the villain, Bolt escapes from his on-set trailer in Hollywood but falls into a box of foam peanuts and unknown to the film company is accidentally shipped to New York City. In New York, Bolt starts to notice that his "superpowers" aren't working, and rationalizes this is the effect that styrofoam has on his body. He then meets Mittens (Susie Essman), a female alley cat who bullies pigeons out of their food. Bolt forces Mittens to help him get back to Hollywood, and after Bolt knocks Mittens unconscious the two start their journey westward on a truck. In Hollywood, Penny is deeply saddened over Bolt's disappearance but is convinced by the studio to continue filming with a Bolt lookalike.

Andy Brewster (Seth Rogen) is a UCLA-graduate organic chemist and inventor. He is attempting to get his environmentally friendly cleaning product, ScioClean, in a major retail store. However, each retail store he visits dismisses him before he can end his pitch. After a disappointing sales pitch to K-Mart, he visits his mother, Joyce Brewster (Barbra Streisand), in New Jersey before leaving on a cross-country trip to Las Vegas, lying to her that his pitch ended well so she won't worry about him. While there she reveals to him that he was named after a boy she fell in love with in Florida named Andrew Margolis, whom she hoped would object to her marriage with Andy's father. However, he never did and she felt that she never mattered to him afterwards. After a little research, he finds Andrew Margolis is still alive and unmarried living in San Francisco. He invites his unknowing mother on the trip, claiming he wants to spend some time with her.

Cal Weaver (Steve Carell) is a middle-aged man who learns that his wife Emily (Julianne Moore) cheated on him with a co-worker, David Lindhagen (Kevin Bacon), and that she wants a divorce. The interaction goes well until they discover that Robbie's teacher is Kate, who reveals to Emily that she and Cal slept together. Cal then confesses to sleeping with many women. Emily leaves in disgust and begins actively dating David. Meanwhile, Hannah, a recent law school graduate, is expecting her boyfriend, Richard (Josh Groban) to propose marriage while they celebrate her passing the bar exam, but he does not, instead, offering her a position at his law firm. Offended and hurt, Hannah returns to the bar where she originally rejected Jacob's advances and kisses Jacob passionately. The two return to Jacob's home to hook up, but end up talking to each other all night and making a connection. Jacob starts a relationship with Hannah, and he becomes distant from Cal.

Long ago, a drop of sunlight became a golden flower capable of healing illness, decay and injury. For hundreds of years the flower is used by Mother Gothel to retain her youth until it is discovered by soldiers of a nearby kingdom. They use the flower to heal their ailing queen, who soon after gives birth to Princess Rapunzel. Gothel discovers that Rapunzel's hair has the flower's healing properties but that attempting to take just the hair by cutting it destroys its power, so she kidnaps Rapunzel and raises her as her own daughter in an isolated tower. The film begins with a woman giving birth to a baby. Within a few minutes of his birth, the baby starts saying: "Ho, ho, ho!". Mother Claus calls her son, Frederick, over to meet his new brother, whom she has decided to name Nicholas. Mother Claus lovingly refers to the new baby as her little Saint Nick. During Christmas, when Nick opens his gifts including Fred's, he decides to give them away to orphans which makes Fred angry. Mother Claus advises Fred to be a better person by stating he should be more like his brother; this in turn causes Fred to resent Nick. Feeling isolated, Fred often climbs a tree to confide his problems in a bird. On one occasion, Nick cuts down the tree, saying that now Fred can bring it inside since he liked it so much. However, the birdhouse Fred's confidant lived in is smashed, causing the bird to leave; this causes a temporary rift between Fred and Nick.

In a world populated by anthropomorphic vehicles, the last race of the Piston Cup championship ends in a three-way tie between retiring veteran Strip "The King" Weathers, infamous runner-up Chick Hicks, and rookie Lightning McQueen. The tiebreaker race is scheduled for one week later at the (fictional) Los Angeles International Speedway in California. Lightning is desperate to win the race, since it would allow him to leave the unglamorous sponsorship of Rust-Eze, a rust treatment for old cars, and allow him to take The King's place as the sponsored car of the lucrative Dinoco team. Eager to start practice in California as soon as possible, he pushes his big rig, Mack, to travel all night long. While McQueen is sleeping, the exhausted Mack drifts off and is startled by a gang of four reckless street racers, causing McQueen to fall out the back of the trailer and into the road. McQueen wakes in the middle of traffic and speeds off the highway to find Mack, only to end up in the run-down desert town of Radiator Springs, and inadvertently ruining the pavement of its main road.
