Robin Wright plays a fictional version of herself, an aging actress with a reputation for being fickle and unreliable, so much so that nobody is willing to offer her roles. Her son, Aaron, suffers from Usher syndrome that is slowly destroying his sight and hearing. With the help of Dr. Barker (Paul Giamatti), Robin is barely able to stave off the worst effects of her son's decline. Robin agrees to sell the film rights to her digital image to Miramount Studios (a portmanteau of Miramax and Paramount) in exchange for a hefty sum and the promise to never act again. After her body is digitally scanned, the studio will be able to make films starring her, using only computer-generated characters.

Two years after their escapade in Las Vegas, Stu Price, Phil Wenneck, Alan Garner and Doug Billings travel to Thailand to celebrate Stu's upcoming wedding to Lauren. Much to Alan's dismay, they are joined by Lauren's younger brother, Teddy. Lauren's father's reveals his disapproval of Stu during a toast. The film opens in a future Paris with scenes establishing the kidnapping of 22-year-old scientist Ilona Tasuiev, who works for the megacorporation Avalon. In the small town of Oakey Oaks, Chicken Little rings the school bell and warns everyone to run for their lives, allowing the whole town into a frenzied panic with much damage ensuing. Eventually, the Head of the Fire Department calms down enough to ask him what's going on and Little explains that a piece of the sky shaped like a stop sign had fallen on his head when he was sitting under the big oak tree in the town square; however, he is unable to find it. His father, Buck Cluck, assumes that this "piece of sky" was just an acorn that had fallen off the tree and had hit him on the head, making Little the laughing stock of the town.

In the year 1962, the Radcliffe family and their one hundred and one dalmatians are preparing to move to their "Dalmatian Plantation," a home in the countryside with plenty of room for all of them. However, one of the puppies, Patch, feels ignored and wishes to be unique like his television hero, Thunderbolt. While watching the Thunderbolt Adventure Hour, he hears about a chance to appear on the show. He is accidentally left behind when his family leaves for the plantation, so he decides to head for the audition to meet his hero and win a guest spot on the show. He fails to impress the producers, allowing a jealous Lil' Lightning to manipulate them into making himself the hero of his own show. Depressed, Patch leaves and runs into Thunderbolt.

In 3028 A.D., humanity has mastered deep space travel and interacted with several alien species. A human invention called "Project Titan" alarms the Drej, a pure energy-based alien species who attack Earth after breaching the Global Defense System. As the Drej start to attack Earth, Professor Sam Tucker, the lead researcher for "Project Titan", sends his son Cale on one of the evacuation ships with his alien friend Tek while Tucker and other members of his team fly the Titan spacecraft into hyperspace. When the Drej mothership destroys the Earth with an energy beam, the surviving humans become nomads, generally ridiculed by other alien species.

The story begins in late 1960s Japan. A group of tanuki are threatened by a gigantic suburban development project called New Tama, in the Tama Hills on the outskirts of Tokyo. The development is cutting into their forest habitat and dividing their land. The story resumes in early 1990s Japan, during the early years of the Heisei era. With limited living space and food decreasing every year, the tanuki begin fighting among themselves for the diminishing resources, but at the urging of the matriarch Oroku ("Old Fireball"), they decide to unify to stop the development.

In 1939 New Orleans, a roguish German Shepherd named Charlie B. Barkin escapes from a dog pound with the help of his friend, a dachshund named Itchy Itchiford. They return to a casino riverboat on the bayou, which is formerly run by Charlie with his partner, a bulldog named Carface Caruthers. Not wanting to share the profits with Charlie, Carface persuades him to leave town with 50% of the casino's earnings. Charlie agrees, but is later intoxicated and murdered by Carface. He is sent to Heaven where a whippet angel named Annabelle (who was named in the sequel) tells him that a gold watch representing his life has stopped. He steals and winds it, sending Charlie back to Earth, but is told that if he dies again, he will go to Hell. After reuniting with Itchy, they discover that Carface is holding an orphan girl named Anne-Marie (presumably kidnapped from the orphanage by Carface), who has the ability to talk to animals and gain knowledge of a race's results beforehand, allowing Carface to rig the odds on the rat races and become rich. They rescue her, intending to use her abilities to get revenge on Carface, though Charlie tells her that they plan to give their winnings to the poor and help her find parents.
