In Chicago, Wesley Gibson (James McAvoy) works at a dead-end desk job with an overbearing boss, takes anti-anxiety medication for panic attacks, and has an unfaithful live-in girlfriend who cheats on him with his friend and colleague Barry. One night at the pharmacy, Wesley is told by a woman named Fox that his recently murdered father was an assassin, and the killer, a man named Cross, is now hunting him. Cross and Fox engage in a shoot-out with hi-tech guns. Wesley panics and runs into the parking lot followed by Cross who steals a truck and attempts to run Wesley down. Fox flips Wesley into her car and she then calmly executes a spectacular escape.

After allowing fugitive Dominic Toretto (Vin Diesel) to evade arrest, former LAPD officer Brian O'Conner (Paul Walker) finds himself on the run from the FBI and leaves Los Angeles to start a new life. He travels across the U.S. in a red Mitsubishi GTO, entering various drag races and winning large amounts of money. The FBI issues a country-wide manhunt against Brian and one day, the police locate him and repossess his car, forcing him to leave on foot. John "Hannibal" Smith (Liam Neeson) is held captive in Mexico by two Federal Police officers working for renegade General Javier Tuco (Yul Vazquez). Hannibal escapes and sets out to rescue Templeton "Faceman" Peck (Bradley Cooper), who is held captive at Tuco's ranch. Hannibal saves Face after enlisting fellow Ranger B.A. Baracus (Quinton Jackson), driving to the rescue in BA's modified GMC Vandura van. Pursued by Tuco, they stop at a nearby Army Hospital to recruit the services of eccentric pilot Howling Mad Murdock (Sharlto Copley). They flee in a medical helicopter, chased by Tuco, in a dogfight that leaves BA with a fear of flying. The battle ends when they lure Tuco's helicopter into American airspace, where it is shot down by a USAF F-22 Raptor for trespassing.

In August 1884, Dan Evans (Bale) is an impoverished rancher and Civil War veteran. He owes money to Glen Hollander (Loftin) and when he fails to pay, two of Hollander's men set his barn on fire. The next morning, as Evans and his two sons drive their herd, they stumble upon outlaw Ben Wade (Crowe) and his gang who are using Evans' cattle to block the road and ambush an armored stagecoach staffed by Pinkerton agents. As Wade loots the stage, Wade discovers Evans and his two sons watching from the hills. Acknowledging that they pose no threat to him and his gang, Wade takes their horses telling Evans that he will leave them tied up on the road to Bisbee.
