I'm Not There uses a nonlinear narrative, shifting between six characters in separate storylines "inspired by the music and many lives of Bob Dylan". Each character represents a different facet of Dylan's public persona: poet (Arthur Rimbaud), prophet (Jack Rollins/Father John), outlaw (Billy McCarty), fake (Woody Guthrie), "rock and roll martyr" (Jude Quinn), and "star of electricity" (Robbie Clark). The film's plot revolves around the coming of the Apocalypse, heralded by the imminent liberation of the Antichrist from the depths of Hell through a certain gateway at the close of one full millennium. This gate can only be opened by a special key, which has been kept guarded by the order of the Templars (who in this version existed since the last days of Jesus). The key in turn is sought out by the servant of the Antichrist, simply known as the Minion, a demonic spirit that transfers itself into the next available host body when his previous one is killed off. His first attempt to gain the key at the close of the year 999 is foiled; the sole surviving Templar of the company charged with hiding the key eventually takes off with a ship to the west, to what was at that time known as the "end of the world", to keep it from the Minion's grasp.

One day, while looking for the family's lost milk cow, Jay Berry discovers monkeys in a nearby river bottom. The storyline follows Morag Gunn from her tough childhood in Manitoba, Canada to middle-age adult life. A key theme of the movie is Morag's search for love. The story also introduces unique characters from the Manawaka series. The film provides a glimpse into many ways that society creates outcasts socially. The film also explores mythology, as illustrated by Christie's Scottish Piper Gunn, Métis hero Jules Tonnerre, Morag's novel and songs by Skinner and Piquette. As Margaret Laurence's crowning achievement, The Diviners shows that truth and love can be "divined" in many ways.

At home, her health deteriorates quickly, resulting in a painful death on the streets before her Manhattan home. In the hospital, it does not take long before Dr. Nora Hart (Jackson) concludes that Sara has died of the Black Death, which has not occurred in centuries. Realizing that the disease is extremely contagious, she tries to push the authorities to warn the New York citizens, but the Mayor is reluctant to cause a widespread panic. Meanwhile, more citizens who have been in direct contact with Sara start to perish. Nora and her new colleague Dr. Jake Prescott (Nordling)—whom she becomes romantically involved with—start a race against the clock to locate and treat all the people who might be infected, while trying to prevent the city from panicking. In the end, Nora is successful in finding everyone who is infected, and treats most of them successfully. Within a week, a pandemic is ended after 22 deaths.
