Kenya McQueen (Sanaa Lathan) is a successful, single African American woman who has sacrificed romance in order to pursue a career as a certified public accountant. Nick Naylor is a handsome, smooth-talking tobacco lobbyist and the vice-president of a tobacco lobby called the "Academy of Tobacco Studies", which for 15 years has been "researching" the link between tobacco smoking and lung cancer. They claim that their research—funded primarily by tobacco companies—has found no definitive evidence of any linkage. Naylor's job consists mainly of reporting the questionable research of the "Academy" to the public and defending Big Tobacco on television programs by questioning opposing health claims and advocating personal choice. Naylor and his friends, firearm lobbyist Bobby Jay Bliss and alcohol lobbyist Polly Bailey, meet every week and jokingly call themselves the "Merchants of Death" or "The MOD Squad".

American Splendor retrace la véritable histoire du scénariste de comics Harvey Pekar. Au début du film, celui-ci mène une existence plutôt morose entre son travail de documentaliste, quelques conversations sur tout et rien avec ses collègues et quelques connaissances, et sa passion pour le jazz, qu'il assouvit en recherchant des pièces rares dans les garage sales (équivalent américain des vide-greniers) ou les magasins d'occasion. C'est ainsi qu'il fait la connaissance du célèbre dessinateur de bande dessinée Robert Crumb, alors inconnu, qui est lui aussi collectionneur. Plusieurs années plus tard, alors que Crumb est devenu célèbre, Harvey lui soumet les story-boards d'un projet de bande dessinée basée sur sa vie, lui proposant d'en assurer le dessin. Crumb accepte, et contre toute attente, cette série intitulée American Splendor relatant une vie simple, banale, obtient un grand succès. Harvey Pekar acquiert ainsi une certaine célébrité aux États-Unis, dont les conséquences sur sa vie quotidienne sont présentées dans le film. En particulier, American Splendor lui permet de rencontrer sa future femme Joyce Brabner, qui écrit elle aussi des comics, et avec qui il va réaliser d'autres histoires autobiographiques, également intégrées en partie dans le film (notamment Our Cancer Year).

The film focuses on Antwone "Fish" Fisher (Derek Luke), a temperamental young man with a violent history who is serving in the U.S. Navy. His father was killed before he was born and his teenage mother, Eva Mae Fisher, ended up arrested soon after and put in jail, where she gave birth to him. He was then placed in an orphanage until such time as she was released and could claim him. Since she had not yet claimed him, at the age of two Antwone was placed in a foster home run by a supposedly religious couple, Mr. and Mrs. Tate (Ellis Williams and Novella Nelson). There, Antwone faced mental and physical abuse by Mrs. Tate for many years until he finally left the home at age fourteen. After living out on the streets for the next few years, he decided to join the U.S. Navy to make something out of his life. However the rough life he had as child caused him to have a violent temper at this point.

Former Army Ranger Cameron Poe is sentenced to prison for manslaughter for using excessive force on a drunk man while trying to protect his pregnant wife Tricia. Poe is paroled eight years later, and is to be released after being flown to Alabama on the Jailbird, a C-123K transport prison aircraft. Along with Poe are several other prisoners including his diabetic cellmate and friend Mike "Baby-O" O'Dell, who is being transferred (but not yet paroled) with Poe. The transfer is being overseen by U.S. Marshal Vince Larkin, as the transfer includes notorious criminal mastermind Cyrus "Cyrus The Virus" Grissom, gangster and Black Guerrilla member Nathan "Diamond Dog" Jones and mass murderer William "Billy Bedlam" Bedford for their transfer to a new Supermax prison. Larkin is approached at the last minute by DEA agents Duncan Malloy and Willie Sims, who ask for Sims to be brought aboard disguised as a prisoner so that he can extract more information from drug lord Francisco Cindino, a prisoner that is to be picked up at Carson City, Nevada en route. Larkin agrees, unaware that Malloy has hidden a gun on Sims' body.
