The story is set during 1907–09 (with an epilogue in 1910), in the Swedish town of Uppsala where Alexander (Bertil Guve), his sister Fanny (Pernilla Allwin) and their well-to-do family, the Ekdahls, live. The siblings' parents are both involved in theater and are happily married until their father, Oscar (Allan Edwall), suddenly dies from a stroke. Shortly thereafter, their mother, Emilie (Ewa Fröling), marries Edvard Vergérus (Jan Malmsjö), the local bishop and a widower, and moves into his ascetic home where he lives with his mother, sister, aunt and maids.

Eva (Liv Ullmann), wife of the village pastor, invites her mother Charlotte (Ingrid Bergman) for a visit to her village. She has not seen her for over seven years. Her mother is a world-renowned pianist, somewhat eccentric, aging, and has survived several husbands. Eva is not as talented as the mother (despite the fact that she has written two books, and has a passing talent at playing the piano). Eva's main concern is to be the mistress of her home, wife, mother and loving sister. It is gradually learned through her dialogue with her mother that her life has had a large number of unfortunate setbacks: a husband (Halvar Björk) she respects, but does not really have affection for, their son drowned when only 4 years old, and Charlotte never appears to have loved Eva as a mother normally loves a daughter. As part of her day-to-day life, Eva takes her disabled and paralyzed sister Helena (Lena Nyman) out of the Hospital into her own home, and she appears to be the only person who can understand her sister's limited speech ability.

A judge in an unnamed country interviews three actors, together and separately, provoking them while investigating a pornographic performance for which they may face a fine. Their relationships are complicated. Sebastian, a volatile heavy drinker in debt, is guilty of killing his former partner and is having an affair with the dead man's widow. Thea, a high-strung woman who is prone to fits and seemingly fragile, is currently married to Sebastian's new partner, Hans. Hans is the troupe's leader. He is wealthy, self-contained, and growing weary of the troupe. The judge plays on the trio's insecurities, but they may have their revenge when they finally perform The Rite in a private session with him.

Persona begins with images of camera equipment and projectors lighting up and projecting dozens of brief cinematic glimpses, including a crucifixion, an erect penis, a tarantula spider, clips from a comedic silent-film reel first seen in Bergman's Prison (depicting a man trapped in a room, being chased by Death and Satan), and the slaughter of a lamb. The last, and longest, glimpse features a boy who wakes up in a hospital next to several corpses, reading Mikhail Lermontov's A Hero of Our Time ("Vår Tids Hjälte" in the film), and caressing a blurry, transient image that shifts between Elisabet and/or Alma's faces.
