Eric Stanton (Andrews), a down-on-his-luck drifter, gets pulled off a bus in the hamlet of Walton because he does not have the $2.25 extra fare to take him to San Francisco. He finds a greasy spoon called Pop's Eats, where Pop (Percy Kilbride) is worried about waitress Stella because she has not shown up for work for days. Ex-New York cop Mark Judd (Bickford) tells him not to worry. Sure enough, the sultry Stella (Darnell) soon returns. Stanton (like the others) is attracted to her, but she is unimpressed by his smooth talk and poverty.

Christopher "Chris" Cross (Edward G. Robinson), a meek amateur painter and cashier for clothing retailer, J.J. Hogarth & Company, is fêted by his employer, honoring him for twenty-five years of dull, repetitive service. Hogarth presents him with a watch and kind words, then leaves getting into a car with a beautiful young blonde. Walking home through Greenwich Village, Chris muses to an associate, "I wonder what it's like to be loved by a young girl." He helps Kitty (Joan Bennett), an amoral fast-talking femme fatale, apparently being attacked by a man, stunning the assailant with his umbrella. Chris is unaware that the attacker was Johnny (Dan Duryea), Kitty's brutish boyfriend, and sees her safely to her apartment building. Out of gratitude and bemusement, she accepts his offer for a cup of coffee at a nearby bar. From Chris's comments about art, Kitty believes him to be a wealthy painter.

"Big John" McMasters (Clark Gable) and "Square John" Sand (Spencer Tracy) are two down-on-their-luck oil wildcatters who join forces. The American government has agreed with the Cherokee Indians to purchase the rights to their land known as the Cherokee Strip (actually, the Cherokee Outlet) in the future state of Oklahoma. After the money arrives by train, it is then loaded onto a stagecoach which subsequently gets robbed by Whip McCord (Humphrey Bogart) and his gang. Jim Kincaid, aka, "The Oklahoma Kid", (James Cagney) sees the robbery, and then ambushes the gang and makes off with the money.
