Vasudevan aka Vasu (Vishal) is a Coimbatore-based moneylender and also was the heir to the Kovai Group, a cloth manufacturing conglomerate, until he was disowned by his mother and matriarch of his joint family Rajalakshmi (Raadhika Sarathkumar) due to some misunderstanding. He meets a rich girl Divya (Shruthi Haasan) at a mall, who soon falls in love with him for his good nature. One day, Vasu saves the Police Superintendent for Coimbatore District Sivakkozhundhu (Sathyaraj) and his wife (Aishwarya) from a group of thugs who work for Anna Thandavam (Mukesh Tiwari), a Pollachi-based businessman and contract killer who kills anyone to acquire their land and money. As Sivakkozhundhu was transferred to Coimbatore district to capture Anna Thandavam, the latter had planned to kill him. Anna Thandavam also plans to illegally grab land sold to a village temple by Vasu's late father. These two incidents cause enmity between Vasu and Anna Thandavam.

After having killed the notorious extortionist Mayil Vaaganam in the first film, Duraisingam (Suriya) has gone undercover after meeting the Home Minister and is working as an NCC officer in a school in Thoothukudi. The only people who know about this operation are the Chief Minister and Home Minister apart from himself. Sathya (Hansika Motwani) is a student who falls for Duraisingam though he is waiting to marry Kavya (Anushka Shetty), his love interest from the previous film. Susai (Santhanam), a local who works in the same school shows Duraisingam parts of Thoothukudi and also tells him about the rogue elements dominating the area helping him in his surveillance. He requests the Home Minister to transfer Erimalai (Vivek) and another cop for assisting him. Sathya's moves are frequently repulsed by Duraisingam who learns that Sathya is the niece of Thangaraj (Rahman). Bhai (Mukesh Rishi) and Thangaraj, are two criminals who control the area. Bhai and Thangaraj have connections with an international gangster and drug lord Danny (Danny Sapani).

David Billa (Ajith Kumar), a Sri Lankan Tamil refugee along with several others, arrives at the town of Rameswaram in boat. After being interrogated by an arrogant civil servant, he is let into the refugee camp where he befriends Ranjith (Yog Japee). David makes his presence felt, when he slaps Muthu (Theepatti Ganesh) one of the rogue workers around for making a derogatory comment. The refugee camp is constantly terrorised by a corrupt police officer Raghubir Sinha (Krishna Kumar). After witnessing the brutal murder of one of the inmates at his hands, David attacks Raghubir when he tries to forcefully detain one of his friends. Raghubir then gets David tortured in prison, and vows to make him suffer. A few days later, Muthu calls David and Ranjith aside and promises them work. Billa and Ranjith are hired to transport fish from Rameshwaram to Chennai. But unknown to them, the truck they were driving had diamonds inside, and Raghubir had alerted his men on the highway to catch them red-handed. As per the plot they get caught in the check post, and after realising what had happened, Billa kills all the cops and drive the truck to its destination. Here they meet Selvaraj (Ilavarasu) who is pleased by Billa's loyalty and includes him in his businesses. Billa gets back and murders Raghubir as well as the arrogant civil servant he had initially met at the camp. He then goes to meet his elder sister (Janaki Sabesh) and his niece Jasmine (Parvathy Omanakuttan) in a church. When they see his gun, they realise that he is involved in crime. Not knowing what to say, Billa simply walks away.

The film begins in Nallur, a small village in Thoothukudi district in southern Tamil Nadu. Duraisingam (Suriya), is the honest police sub inspector of Nallur, assisted by his bumbling colleague Erimalai (Vivek). He belongs to Nallur with his father having a respectable status in the village. His family business is provision stores and Duraisingam wants to join it, but he joined police due to his father's wishes. He resolves most of the problems in his village with non-violence and mutual counselling. He uses force only when the situation demands it, thereby gaining much reputation and love from the villagers. Mahalingam (Nassar) is an industrialist in Chennai and a friend of Singam’s father (Radha Ravi). He comes to the village with his beautiful daughter Kavya (Anushka Shetty). Eventually Singam and Kavya fall in love with each other.

Deva (Suriya) arrives at the Chennai International Airport after running an errand for Arumuga Dass (Prabhu Ganesan) of smuggling pre-release pirated movies on DVD. The two leave for their hideout and instruct Dilli (Karunas), a hearing-impaired assistant of Dass, to make copies of the DVD. Just when police crews arrive into their vicinity, Dass informs Deva and the rest to leave. Deva tells the others to leave while he quickly sets up the burning process. Deva also leaves the hideout just in time. However, the police officers arrive at the hideout, only to see the burning process of the DVDs complete. The police then seize the DVDs and computers. The inspector, who was a friend of Dass, tells him that he can close the case if one of Dass's men agrees to the crime. Deva attempts to go, but another man, Chitti (Jagan), who had no affiliation to them volunteers, takes the blame. Later, Chitti joins Dass's group.

Gautham Subramaniam (Jayam Ravi) is a doctor from an Indian family who lives in Chennai. Since his father (Nizhagal Ravi) and mother are doctors, he wanted to become a doctor. Gautham gets an opportunity from the Government of India to go to Russia to represent India in a week long medical conference, along with five other young doctors. Gautham is delighted to receive this offer and packs his bags to fly to Russia. Meanwhile, his beloved is introduced on the frame which happens to be Shenba (Kangana Ranaut). She happens to be the same village girl in which Gautham's sister Sarasu, (Anu Haasan) resides with her husband (Chetan) and so they two meet and fall in love. Despite initial frictions between her father and uncle (Mahadevan and Bose Venkat) with Gautham, they eventually agree to get the pair married. Gautham’s parents agree to the marriage and as a happy man Gautham is ready to marry Shenba after his return to India from his Russia trip.

Vallavan (Silambarasan) is a happy-go-lucky college student with a set of friends. Bala (Santhanam) & Suji (Sandhya) are two of Vallavan's friends. Rani (Bidushi Dash Barde), the daughter of former Chennai police officer Arokiya Raj (Prakash Raj), tells her father over the phone that she will back home in 20 minutes. As she leaves the telephone booth, she is approached by someone. The screen then cuts to black and we find out that Rani does not come home. The following morning Arokiya Raj finds a finger hanging at his door. Police reports confirm that it is his daughter Rani's finger. He calls Raghavan (Kamal Hassan), his old friend, for investigation. Raghavan handles the case and they eventually find the body of Rani in a forest in the city outskirts. The doctor's report states that the killer bisected Rani’s body with a surgical knife, proving that the murderer has a strong medical background. Following the trauma of losing their only daughter, Arokiya Raj and his wife move to New York.

Shiva falls in love with her but soon realises that their characters are totally different. Pooja feels alone, like a fish out of water in India and she wants to return to the US. So Shiva thinks about a plan to get her to stay and he expresses his love to her by creating a fictitious character MR X but never reveals about the character till the end. Pooja(Renuka Menon) who got impressed by MR X refuses to see him in the later stage but understand and accepts shiva's true love.

One day, Meera and Arjun get into a quarrel on the road and Meera gets into an auto. Arjun hitches a ride from Michael, who is riding in the same direction, to catch up with Meera. Suddenly, Michael is shot but Arjun saves him. After talking to Michael, Arjun changes his mind and wants to be a politician. Selvanayagam is worried when he hears news of students standing in the election. He uses every possible resource to get them out of Politics. Firstly he provides scholarship to study at a prestigious foreign university to Michael. When Michael refuses the bribe, he orders his goon Inba to take control. Inba beats some students but faces very strong retaliation from Michael and other students. He kidnaps Arjun and other candidates. However, they manage to escape with the help of Inba's ally, Dilli (Sriman). Inba follows Arjun and beats him up. While running, Arjun manages to call Michael for help. He comes to rescue Arjun at Napier Bridge. Then Inba is handed to the police. Michael wins the four seats he and his college mates have contested for. Sashi leaves for her hometown while Inba remains in prison hoping not to get the capital punishment. Michael, Arjun and two friends enter into politics.

Munna (Siddharth Narayan), Babu Kalyanam aka Bob Galy (Bharath), Juju (Nakul), Krishna (Thaman) and Kumar (Manikandan) are five friends. They come from different backgrounds – Munna from a middle-class family, Bob Galy from a Telugu-speaking family, Juju from an academic-achieving family, Krishna from a rich family and Kumar from a poor family. Much to the chagrin of their parents, they have no interest in studies. They meet a depressed middle-age man, Mangalam (Vivek) in a bar one day and help him home after he passes out. Although they first use him as a source to get alcohol, Mangalam enjoys their company and becomes a mentor of sorts for the boys.They even go as far as hiring a prostitute(whose contact was obtained from mangalam) to have sex with while Munna's parents are out of station (Bhuvaneswari), but all of them back down at the last moment.

Sridhar (Tarun) is a final year degree student and gets selected in the campus selections for studies abroad. He goes to Mumbai for an interview and in his return finds Preeti (Trisha) and loses his heart to her. Sridhar comes to know that Preeti was about to join in degree and Preeti learns that he was a final year degree student. Sridhar is an ardent cricket fan, so is Preeti, while Sridhar is also a good football player. The principal of the college appoints a coach called Reshma (Shriya), who is also a student, for the football team led by Sridhar. Sridhar tries his best to trace Preeti by visiting several women's colleges, while Preeti also watches for Sridhar at the men's college. Accidentally, Preeti helps Sridhar's mother, when she had a heart attack. In order to find Preeti at any cost, Sridhar goes to the LB Stadium, where a cricket match is in progress and gets injuries in a bomb blast in the stadium. Just before the blast, both Preeti and Sridhar find each other but could not meet due to the blast.
