In 1996, Jamie Randall (Jake Gyllenhaal) is fired from a Pittsburgh electronics store for having sex with his manager's girlfriend. His wealthy brother Josh (Josh Gad) announces at the dinner table at their parents' (George Segal and Jill Clayburgh) house that he has found Jamie a job as a pharmaceutical sales representative. After attending a Pfizer training program where he has sex with the instructor (Kate Jennings Grant), Jamie goes to work for the company and tries to get doctors to prescribe Zoloft and Zithromax. He is rebuffed, much to the dismay of his regional manager, Bruce (Oliver Platt), who sees Jamie as his ticket to the "big leagues" of Chicago. Bruce says if Jamie can get Dr. Knight (Hank Azaria) to prescribe Zoloft instead of Prozac, other doctors will follow his lead. Jamie tries to get access to Dr. Knight by hitting on his female employees until, exasperated, Dr. Knight unethically permits him to observe him at work, during which time he accidentally sees a disrobing patient, Maggie Murdock (Anne Hathaway), who suffers from early onset Parkinson's disease.

Young filmmaker and scientist Victor Frankenstein lives with his parents, Edward and Susan Frankenstein, and his beloved dog, Sparky, in the quiet town of New Holland. Twenty-seven-year-old Howard the Duck (Chip Zien) lives on Duckworld, a planet similar to Earth but inhabited by anthropomorphic ducks and orbited by twin moons. As he is reading the latest issue of Playduck magazine, his armchair begins to quake violently and propels him out of his apartment building and into outer space; Howard eventually lands on Earth, in Cleveland, Ohio. Upon arriving, Howard encounters a woman being attacked by thugs. He defeats them using a unique style of martial arts. After the thugs flee, the woman introduces herself as Beverly Switzler (Lea Thompson), and decides to take Howard to her apartment and let him spend the night. The following day, Beverly takes Howard to Phil Blumbertt (Tim Robbins), a scientist who Beverly hopes can help Howard return to his world. After Phil is revealed to be only a janitor, Howard resigns himself to life on Earth and rejects Beverly's aid. He soon applies for a job as a janitor at a local romance spa. Because of unfair treatment by his boss, Howard resigns and rejoins Beverly, who plays in a band called Cherry Bomb. At the club at which Cherry Bomb is performing, Howard comes across their manager (Tommy Swerdlow), and confronts him when he insults the band. A fight breaks out, in which Howard is victorious.

In 1952, Ed Wood (Johnny Depp) is struggling to join the film industry. Upon hearing of an announcement in Variety magazine that producer George Weiss (Mike Starr) is trying to purchase Christine Jorgensen's life story, Ed is inspired to meet Weiss in person. Weiss explains that Variety 's announcement was a news leak, and it is impossible to purchase Jorgensen's rights. The producer decides to "fictionalize" the film, titled I Changed My Sex!. Ed tries to convince Weiss that he is perfect to direct I Changed My Sex! because he is a transvestite, but is unsuccessful since Weiss wants a director with experience. One day, Ed meets his longtime idol Bela Lugosi (Martin Landau), after spotting him trying out a casket. Ed drives Bela home and the two become friends. Later, Wood persuades Weiss to let him direct I Changed My Sex! by convincing him that having a star in the film would sell tickets, and they could sign Bela for a low price.
