Henry Altmann (Robin Williams) is stuck in Brooklyn traffic while on his way to a doctor's appointment. His car is suddenly struck by a taxi, which propels him into a rage which he unleashes upon the taxi driver. Henry goes to the Brooklyn hospital, where Dr. Sharon Gill (Mila Kunis) is covering for his usual doctor, with whom she is having an affair. After examining scans of his brain, Sharon informs Henry that he has a brain aneurysm and his outlook is bad. Henry erupts, throwing insults at Sharon and demanding that she tell him how long he has to live. She tries to dodge the question, but Henry is persistent. Panicking, Sharon tells him that he will die in ninety minutes. Henry leaves the hospital in a rage.

Abel Morales (Oscar Isaac) is the owner of Standard Heating Oil Co, an up-and-coming company. The company has been plagued by the hijacking of several trucks, each carrying thousands of dollars in heating oil. One young driver, Julian (Elyes Gabel), is severely beaten when his oil truck is hijacked by two unknown assailants. Abel's wife, Anna Morales (Jessica Chastain), beseeches Abel to fight violence with violence, but Abel refuses. The story follows the characters of Molly (Bledel), a struggling waitress, and Gus (Levi), a jewelry store employee. Gus and Molly met through a chance encounter when Gus is at Molly's diner ordering a coffee. Molly sees Gus making notes in his pen recorder and when he goes round back to take a phone call leaves a message on it saying her name. Molly and Gus then begin to date, and it is learned that Gus has a memory defect where he can not make new memories and forgets the day every time he sleeps. Trouble ensues as he does not tell Molly this and she finds him recording all their meetings. Gus tries to explain, but Molly mistakes this for being odd. A few weeks later and Molly returns to the apartment to find Gus's sister who explains and gives her the scrapbook Gus made entitled "Molly" he made to remember her. Molly visits the hospital where Gus Is under observation and introduces herself as his girlfriend and gives him the scrapbook so he can remember their relationship. We learn that Gus was meant to be the next Einstein working at Nasa in California, before his accident. Molly takes Gus there, where he meets old colleagues who tell him his work will help understand the universe much better. Whilst there Molly meets Gus's ex girlfriend who warns her any serious relationship with Gus will mean he won't remember their kids and other events in the long-term, and Gus also tells her to leave him if an operation to cure his disease does not work. Gus's sister tells Molly that Gus has already had this operation, however, but that it did not work. That night Molly sees a note on Gus's laptop for his to do list for the next day "Ask Molly to marry me". Molly deletes the note as a result of what Gus had told her to do and Gus wakes up the next day having never remembered Molly. Molly continues her life with a quarter million pounds she got from her great Aunt's will and starts a flower business. One day, at a wedding, a shooting star appears and Molly is reminded of Gus. She visits him the next day and hints to Gus that they know each other and asks him for coffee. The movie ends.

18-year-old Harry (Norman Reedus), is an innocent, bashful burger boy who lives with his overly attentive mother Kate. Harry's father left the family sometime before the events of the film, leaving Kate for another man. They live in a ratty old apartment, where Kate treats her son like a child, even going as far to draw his bath water and connect a wire to his reading lamp, shutting it off when Harry is busy to get his attention. One day, Arnie (Adrien Brody), Harry's oldest and best friend, goes to a strip club, where the boss owes Arnie's mob boss money. Harry watches as Arnie beats the owner, and snaps, releasing his rage out on the owner, pummeling him to the point where Arnie has to pull Harry off to avoid killing him. Outside, both are visibly shocked by Harry's outburst, but Harry is shocked and confused at the fact that he liked it. Not long after, Arnie's boss Abie Pinkwise, meets the two at a local diner, where he remarks how much potential Harry has in the mob business. He invites Harry to become his apprentice, and Harry accepts. After leaving, Arnie attempts a heist at a small store, but backfires when the clerk holds him at gun point, sending him to jail. When Harry is told to ditch a car (evidence in a homicide), by his bosses, he leaves evidence (a magazine with his name on it), and a witness. He is arrested but is proven to be loyal to his employers by keeping silent, despite being beaten by the police. Now trusting him, the bosses get him out of jail and take him out to celebrate at a brothel. When alone with one of the women, Harry is un-aroused, and timidly asks the prostitute if he seems normal. When he answers that he doesn't feel that way for neither men nor women, she gently replies that she isn't the kind of professional he should be talking to. He goes home dejected, and when his mother smells perfume on him the first signs of her jealous tendencies begin to show.

Michael Makeshift (Jason Alexander) is a schnook who can't get over the fact his girlfriend dumped him. To make matters worse, his landlord is beginning to bug him for long-overdue rent. Things for Michael's brother Reggie (James Woods) are much better. He is planning to rob the credit union where his own mother (Bea Arthur) works. His new wife Valerie (Lolita Davidovitch) has no idea what Reggie is planning and is therefore puzzled when she awakes at Michael's apartment after Reggie left her there whilst she was drunk and unconscious. She also does not know that Reggie has paid Michael for the favor. Despite his well-laid plans, things go badly for Reggie when his henchmen mutiny and force him to reveal that the clueless Valerie carries the security codes they need to pull off the job in her suitcase, which in turn begins a merry chase. While Michael helps Valerie escape, he reveals the truth about Reggie. At the same time, she becomes increasingly attracted to her new protector.
