Akeem Joffer, the crown prince to the throne of the wealthy African nation of Zamunda, lives a pampered lifestyle with every daily facet performed by servants. Akeem has become fed up with this and wishes to do more for himself. The final straw comes when his parents, King Jaffe and Queen Aeoleon, present him with an arranged bride-to-be named Imani Izzi, whom he has never met and who has been trained to obey Akeem's every command. Akeem concocts a plan to travel to the United States to find an intelligent, independent-minded woman he can both love and respect, and who will love Akeem for who he is and not for his wealth and social status as a prince. Akeem and his best friend/personal aide, Semmi, flip a coin to decide between going to either Los Angeles or New York City, and end up going to New York City. They end up in the borough of Queens and rent a run-down apartment in the neighborhood of Long Island City, passing themselves off as poor foreign students. They begin working at a local fast food restaurant called McDowell's—an obvious ripoff of McDonald's—owned by widower Cleo McDowell and his two daughters, Lisa and Patrice.

One night in Detroit, Axel Foley (Eddie Murphy) plans to arrest a gang of car thieves who run a local chop shop. Unbeknownst to his boss, Inspector G. Douglas Todd (Gil Hill), Foley has canceled the SWAT team, intending to raid the shop using only his team. Meanwhile, a group of men arrive at the chop shop to pick up a cube van that the car thieves had hijacked. The leader of the group confirms that the vehicle still contains its cargo, which consists of crates labelled as property of the U.S. government, then has his men execute the car thieves.

Duke brothers Randolph (Ralph Bellamy) and Mortimer (Don Ameche) own a successful commodities brokerage in Philadelphia. Austin Millbarge is a geekish, basement-dwelling codebreaker for The Pentagon who aspires to escape his under-respected job to become a secret agent. Emmett Fitz-Hume, a wisecracking, pencil-pushing son of an envoy, takes the foreign service exam under peer pressure. Millbarge and Fitz-Hume meet during the test, on which Fitz-Hume openly attempts to cheat after an attempt to seduce his immediate supervisor in exchange for the answers backfires. Millbarge, however, was forced to take the test, having had only one day to prepare after his supervisor gives him a notice that was two weeks old.

The film portrays two conflicts that take place around Flagstone, a fictional town in the American Old West: a land battle related to construction of a railroad, and a mission of vengeance against a cold-blooded killer. A struggle exists for Sweetwater, a piece of land near Flagstone containing the region's only water source. The land was bought by Brett McBain (Frank Wolff), who foresaw that the railroad would have to pass through that area to provide water for the steam locomotives. When crippled railroad tycoon Morton (Gabriele Ferzetti) learns of this, he sends his hired gun Frank (Henry Fonda) to intimidate McBain to move off the land, but Frank instead kills McBain and his three children, planting evidence to frame the bandit Cheyenne (Jason Robards). It appears the land has no owner; however, a former prostitute (Claudia Cardinale) arrives from New Orleans, revealing she is Jill McBain, Brett's new wife and the owner of the land.

Link Stuart (Bronson) is a ruthless outlaw, and co-leader along with Gauche (Delon) of a gang of bandits. Link and Gauche lead their gang on a successful train robbery, and discover that one of its cars carries a Japanese ambassador, who is bringing a ceremonial katana (sword) as a gift for President Ulysses S. Grant. Gauche takes the sword, and kills one of the two samurai guards, while members of his gang attempt to murder Link by throwing dynamite into the train car he occupies, then leaving him for dead.

The film opens as the half-Apache Chato orders a drink at a bar. The bartender ignores him and serves the local sheriff who has arrived after Chato. The sheriff calls Chato a "redskin" and tells him the bar is for whites only. He moves behind Chato while hurling a stream of abuse at him. The sheriff's taunts escalate, and he draws his gun while saying that he is going to kill Chato. Chato whirls around and shoots the sheriff in the gut, killing him. He rides out of town on his Appaloosa.
