Mike Enslin (John Cusack) is a cynical, skeptical author who, after the death of his daughter Katie (Jasmine Jessica Anthony), writes books appraising supernatural events in which he has no belief. After his latest book, he receives an anonymous postcard depicting The Dolphin, a hotel on Lexington Avenue in New York City bearing the message, "Don't enter 1408." Viewing this as a challenge, Mike forces the hotel to allow him to book the room, referencing a law that any hotel room in New York can be requested as long as it meets safety standards. The hotel manager, Gerald Olin (Samuel L. Jackson) tries to dissuade Mike from checking into the room, explaining that 56 people have died in the room over the past 95 years, and that no one has lasted more than an hour inside it. Mike, who does not believe in the paranormal, insists on staying in the room, and asks Olin if he thinks it is haunted; Olin replies that it is "evil" rather than haunted.

The film is set in England where an American pathologist Dr. Richard Murray is grieving for his late wife Carol who died in a car crash a few months previously. Five years after the first film, Crown Princess of Genovia Amelia "Mia" Thermopolis (Anne Hathaway) has just graduated from Princeton University's Woodrow Wilson School and is returning to Genovia with her bodyguard Joe (Héctor Elizondo) and beloved cat Fat Louie. There, she will await her reign once her grandmother, Queen Clarisse (Julie Andrews), steps down as Queen. During Mia's 21st birthday party, she dances with all the eligible bachelors in hope of finding a husband. She becomes attracted to a handsome young gentleman named Nicholas (Chris Pine). During the course of the night, Mia's tiara falls off and is caught by Parliament member Viscount Mabrey (John Rhys-Davies) who secretly plans to steal Mia's crown. While the Parliament is in-session the next morning, Mia stumbles upon a hidden room that allows her to secretly listen in. Viscount Mabrey reveals his nephew, Lord Devereaux, is another heir to the Genovian throne. Despite Queen Clarisse's objection, the only way Mia can assume her duties as Queen is if she marries within the month. Clarisse invites Lord Devereaux to stay at the palace, while Mia is shocked to discover Lord Devereaux is Nicholas. Mia's best friend Lilly Moscovitz (Heather Matarazzo) surprises her by visiting. Together, they pick through potential husbands. Mia eventually chooses Andrew Jacoby (Callum Blue), Duke of Kenilworth and days later they are engaged. Mabrey plans to have Nicholas woo Mia and dissolve the engagement.

A former CIA agent and ex-Special Forces member Dean Cage (Wesley Snipes), is in a rehab program, haunted by a botched mission in Bosnia which resulted in the execution of his best friend Scott (Cristian Solimeno). Abelard is a famous teacher of philosophy at the cathedral school of Notre Dame, and a champion of reason, at a time when academics are required to observe chastity. It's New Year's Eve, Johnny Reeve (Karl Howman) and his mate, Toby (Daniel Peacock) are out cruising in South London being followed by three quasi-threatening motorcyclists.
