Two years after their escapade in Las Vegas, Stu Price, Phil Wenneck, Alan Garner and Doug Billings travel to Thailand to celebrate Stu's upcoming wedding to Lauren. In 1954, a nuclear bomb is detonated at the moment a giant creature emerges from the ocean. In 1999, Project Monarch scientists Ishiro Serizawa and Vivienne Graham investigate a colossal skeleton unearthed in a collapsed mine in the Philippines. They find two spores; one dormant and one hatched that made a trail into the sea. In Japan, the Janjira Nuclear Power Plant experiences unusual seismic activity and Supervisor Joe Brody sends his wife Sandra and a team of technicians into the reactor. A tremor breaches the reactor, leaving Sandra and her team unable to escape while the plant collapses.

To celebrate his upcoming marriage to Tracy Garner (Sasha Barrese), Doug Billings (Justin Bartha) travels to Las Vegas with his best friends Phil Wenneck (Bradley Cooper) and Stu Price (Ed Helms), as well as Tracy's brother and Doug's future brother-in-law Alan (Zach Galifianakis), in Doug's future father-in-law's vintage Mercedes-Benz. They stay at Caesars Palace, where they relax in the room before celebrating with a few drinks on the hotel rooftop. Two years after the events in Bangkok, Leslie Chow (Ken Jeong) escapes from a maximum security prison, using a riot as cover. Meanwhile in America, Alan Garner (Zach Galifianakis) causes a 20-car freeway pileup after he purchases a giraffe and accidentally decapitates it on a low overpass. Alan's father Sid (Jeffrey Tambor), angry with Alan for his immature antics and never owning up to his mistakes, dies of a heart attack in the middle of a lecture. After the funeral, Alan's brother-in-law Doug Billings (Justin Bartha) informs friends Phil Wenneck (Bradley Cooper) and Stu Price (Ed Helms) that Alan has been off his ADHD medication and is out of control. They attend an intervention, in which Alan agrees to visit a rehab facility in Arizona, so long as "the Wolfpack" takes him there. Peter Highman (Robert Downey, Jr.) is on a plane, flying home to be with his wife Sarah (Michelle Monaghan), who is due to give birth. Graeme Willy (Simon Pegg) and Clive Gollings (Nick Frost) are British comic book enthusiasts and best friends who travel to the United States to attend the annual San Diego Comic-Con International, and to take a road trip through the American Southwest to visit sites significant to UFO lore.
