The film is a celebration of the casting profession, highlighting its previously unsung role in film history while also serving as an elegy to the lost era of the New Hollywood. It focuses on casting pioneer Marion Dougherty, an iconoclast whose exquisite taste, tenacity and gut instincts brought a new kind of actor to the screen that would mark the end of the old studio system and help to usher in this revolutionary new period. The film draws a line through the last half century to show us the profession’s evolution from studio system type-casting to the rise of large ensemble films populated with unique and diverse casts.

Set amidst Chicago's swing music revival of the late 1990s, Oscar Novak (Perry), an aspiring architect and his business partner, Peter Steinberg (Platt), have just landed a career-making opportunity with a Chicago tycoon Charles Newman (McDermott) who has chosen them to compete for the design of a multimillion dollar cultural center. In a ploy for publicity, Newman has pitched Oscar and Peter in a neck-and-neck competition with their archrivals and former colleagues, the hugely successful (and equally ruthless) Decker and Strauss. In a comic twist Oscar is mistaken for a gay man when meeting with Charles Newman (made even more humorous by the fact that Peter is genuinely gay, with Oscar's comments leading Newman to think that Peter's the straight one). Under the mistaken impression that Oscar is homosexual, and therefore a safe companion for his girlfriend Amy (Campbell), he asks Oscar to keep an eye on her for him and make sure that she doesn't talk to his wife. Oscar falls for Amy virtually on sight, but she thinks he's gay. He is forced to maintain the charade to avoid getting into trouble with Newman, and losing the commission.

Lorna Cole (Rene Russo) is pregnant with LAPD sergeant Martin Riggs' baby; they are not married, but both are thinking about it. Police sergeant Roger Murtaugh's daughter Rianne (Tracie Wolfe), is also pregnant, and Riggs later learns from Lorna that Rianne has secretly married Lee Butters (Chris Rock), a young detective who works at the same station as Murtaugh and Riggs – secretly because Roger had made it clear that he does not want his daughter to marry a police officer. Due to the many disasters caused by both sergeants while on duty, the police department has lost its insurance carrier, and cannot get a new one while they're still out on the streets. Because firing them is not an option, the Chief uses a special privilege and they get temporarily promoted to captains, given that there aren't any lieutenant spots available.

L.A.P.D. Sergeants Martin Riggs (Gibson) and Roger Murtaugh (Glover) arrive at an evacuated building believed to have a bomb in it. Against orders, they go inside to investigate the bomb. Riggs decides to deactivate the bomb himself instead of waiting for the Bomb Squad to arrive. Unfortunately, he causes the bomb to detonate and the whole building to collapse. This causes them to be demoted to uniform duties, which upsets Murtaugh because he only has seven days until retirement. While on street patrol, the two thwart a robbery using a duplicate armored car, although they only manage to catch one of the two would-be thieves. During the arrest, it is discovered the criminals were armed with a new type of armor-piercing bullet, informally referred to as "cop killers". For their efforts in stopping the raid, Riggs and Murtaugh are reinstated as Detective Sergeants by their superior, Captain Murphy (Kahan). Murphy introduces internal affairs Sergeant Lorna Cole (Russo) to the two detectives and informs them that the suspect they have in custody from the raid is a known associate of former L.A.P.D.
