Thomas "Tom" Popper (Jim Carrey) is a divorced real-estate entrepreneur whose father traveled around the world during his childhood. He learns that his father has died during an adventure to Antarctica and discovers that his father's will asks for his son to be given a souvenir from his last adventure. The next week, a crate containing a gentoo penguin (Captain) shows up at his doorstep. Eventually, five more penguins arrive. Popper intends to give them away but changes his mind when his children, Janie and Billy (Madeline Carroll and Maxwell Perry Cotton), think that the animals are Billy's birthday present. That night, Popper meets with New York zookeeper Nat Jones (Clark Gregg), who asks for the penguins. Popper tries to stall him by asking him to collect the penguins another time. Jones agrees but not before warning that the conditions in Popper's apartment are not good enough to raise penguins. Popper is forced to pay the board of his apartment to keep the penguins in his apartment and lies to his arrogant neighbor Kent (David Krumholtz) about there being no animals in his home.

Anna Coleman (Lindsay Lohan) is an average teenager who constantly fights with her stodgy widow mother Tess (Jamie Lee Curtis) and annoying younger brother Harry (Ryan Malgarini). The two often fight about Anna's rock band which Tess hates because she thinks that they are all noise, Tess's upcoming second marriage to Ryan (Mark Harmon) which Anna is not emotionally ready for because her father died three years ago, and Anna's claims about her archrival Stacey Hinkhouse (Julie Gonzalo) who doesn't stop torturing her and has Tess convinced that she and Anna are still best friends like they used to be and sadistic English teacher, Mr. Bates (Stephen Tobolowsky) who gives her an "F" on everything she does no matter how hard she tries. Sixteen-year-old homeschooled Cady Heron (Lindsay Lohan) and her zoologist parents (Ana Gasteyer and Neil Flynn) return to the United States after a 12-year research trip in Africa, settling in Evanston, Illinois. Cady attends a public school for the first time, meeting new classmates Janis (Lizzy Caplan) and Damien (Daniel Franzese). The two warn Cady to avoid the school's most exclusive clique, the Plastics, who are led by queen bee Regina George (Rachel McAdams). The Plastics take an interest in Cady, however, and start to invite her to sit with them at lunch. Seeing that Cady is slowly becoming one of The Plastics, Janis hatches a plan of revenge against Regina, using Cady as the infiltrator.
