Scientist Rohit Mehra lives with his son Krishna "Krrish" Mehra (both played by Hrithik Roshan) in Mumbai. Krishna is married to Priya (Priyanka Chopra) who works as a journalist for Aaj Tak. Rohit works in a research institute for the Indian government and Krishna, while moon lighting as the superhero Krrish, is regularly fired from various day jobs due to lack of attendance. Rohit is researching on a device that will bring life to dead tissues by the use of solar energy. Despite several attempts, he fails to complete the experiment due to the high intensity of sun rays. On a usual day, Krishna loses his job from the security wing of a company when he hears news about an aeroplane whose front tyres are stuck due to some malfunctioning which creates high risk, even if landed, it may explode and so will the passengers inside. Krishna immediately changes himself to Krrish and saves the plane from accident by holding the component to make tyres free to roam.

The film starts with ACP Yashvardhan IPS (John Abraham) in the hospital, recalling the accident that changed his life forever. Yash is a dutiful, responsible and stone-hearted police officer in the Narcotics Control Bureau whose only passion in life is crime-fighting. He has no family or loved ones and has been alone in life since he can remember. Everything changes when Yash meets Maya (Genelia D'Souza), an independent and lovely woman. Though the two get off to a rocky start, Maya warms up to him once she realises he is a police officer, and apologises for the misunderstandings they had. When Maya gets hit by a car in front of Yash, he rushes her to the hospital. With that, the two bond and start to meet and go out regularly. Maya confesses her love to Yash, but Yash refutes her advances, as he does not want her to become his weakness in his professional life. Madhur Chaudhary (played by Mohnish Bahl) moves out of his rural home leaving his father alone after various disputes they have. Madhur re-locates to Delhi along with his wife, Komal, and daughter, Ria (played by Yuvika Chaudhary). Sixteen years later, Ria is now in college, while Madhur is an Investigator with Human Rights Department. He is asked to investigate a case in his village of alleged suicide of a couple, Veer and Surili. The locals, including the fathers of the couple, Daulat Singh and Sukhiram respectively, admit that the couple had killed themselves. After going in depth in his investigation Madhur finds out that the deaths are one of many that have taken place in a region that is still bound by Khap Panchayat. It dictates that couples cannot marry distant relatives nor can they have an inter-caste/inter-religious marriage. As he delves further into this issue, he ends up being attacked, is hospitalized and then subsequently dies. Komal and Ria move in with Madhur's father Omkar Singh Chaudhary (played by Om Puri) and eventually settle down in the village.

Sameer Behl (Shahid Kapoor) comes to Mumbai with Bollywood dreams, struggles through the day as a courier boy and keeps failing in auditions for advertisements. More stereotypes are stuffed in as he refuses to take help from his dad in Delhi, is thrown out from his rented flat by the landlord and comes away with his calculative companion. One day, a movie director, Rajeev Sharma, (Mohnish Behl) sees Sameer dancing and calls him to his office. Soon Sameer is signed as the male lead in his film and friend Tina (Genelia D'Souza) is selected as the choreographer on the same project. Homeless, Sameer sleeps in his car and works as a dance teacher in a school. Subsequently, Sameer loses his film and is disillusioned. Tina finds this out and asks him how he can spend his life like this. After some time, Tina tells him that she also quit the film and did not want to be the choreographer for that film because she saw him upset. Sameer realised the love Tina had for him. Tina encourages him to participate in a television talent-hunt show, the winner of which will win the same role that Sameer was to play. After some time he agrees to participate in it. After filling up the contest form, he also says "I love you" to Tina. In his screen-test, Sameer dedicates his monologue to his mother. After his monologue, during the interview, he comes to know that his father's shop in Delhi has been demolished. He promises Tina that he will come back for her and leaves. He leaves Mumbai to meet his father, heart-broken about the whole thing. After watching Sameer's performance in the TV, his father convinces him to go back to Mumbai and participate in the show.

The story begins in Ajmer, in the state of Rajasthan, where Rajnandini Khandelwal (Sandeepa Dhar) passes her 12th class examination and is the state-topper, winning three scholarships. Everyone is happy, but in keeping with her father's conservative tastes, he and her mother want Rajnandini to get married. Vijay Harshwardhan Malik (Amitabh Bachchan), the ethical CEO of struggling television channel India 24/7, is losing the ratings battle with a rival channel headed by Amrish Kakkar (Mohnish Behl). Malik's son Jai (Sudeep) makes a deal with a wealthy and corrupt politician, Mohan Pandey (Paresh Rawal), to frame the Prime Minister (K K Raina) for being complicit in a terror attack. Pandey can then take over the position and Jai will have enough money to start his own channel. Jai's brother-in-law (Rajat Kapoor) supports Mohan Pandey because he wants to become the biggest industrialist in the country and Pandey could help him by framing such policies. Jai shoots a short video featuring his friend Khanna and a close friend of the PM. This meeting is staged and the PM's friend, under duress, says that the PM was involved in plotting a bomb blast so as to create fear and panic among the people so that he could get a bill passed. Jai convinces his father that the story is true and believing it to be so, Malik airs it on his network. The scandal rocks the nation and elections are held in which Mohan Pandey wins. He becomes the PM. However, one of Malik's reporters, Purab (Ritesh Deshmukh), discovers the plot. He initially approaches Amrish Kakkar with a request to air his findings on the news. Amrish, though, cuts a deal with Mohan Pandey and does not air the CD. Purab then tells his boss of his findings on the day his son is getting engaged. Malik goes on air one final time and confesses the wrongdoings of his son, son-in-law and exposes Mohan Pandey. Unable to bear the guilt, Jai commits suicide. Mohan Pandey denies his role in the scandal. Malik steps down as the CEO of the news channel and hands over the baton to the reporter who exposed the truth.

After receiving a generous grant from the American Medical Association Dr Duke Chawla (Bobby Deol) who always gives hope to his patients, proposes to his sweetheart, Pooja (Kangana Ranaut), and both arrange to get married soon.
