At the beginning of the film, we learn that the story takes place in one of many alternate worlds, in which a person's soul is contained within an animal companion called a dæmon (pronounced demon). The Magisterium, represented as a unified religious power, exercises power in the secular world. Lyra Belacqua, an orphan that resides in Jordan College, in a city that resembles Oxford, with her dæmon Pantalaimon (Pan), accidentally witnesses a Magisterium member poison her uncle's bottle of Tokay. Lyra then warns her uncle, Lord Asriel, who instructs her to remain in hiding. Lyra watches Asriel give a presentation regarding Dust, a particle that the Magisterium has forbidden the mention of. The college gives Asriel a grant to fund a northern expedition.

Gaylord "Greg" Focker (Stiller) is preparing to celebrate his twins' fifth birthday party. Things seem to go awry when Greg's father-in-law Jack Byrnes (De Niro) visits. Recently, Jack has been diagnosed with a heart condition and become embittered by his daughter Debbie's divorce from her husband, Bob (whose marriage was the social event in the first film and how Jack and Greg met), for cheating on her with a nurse. Jack's plan was originally to name Bob as his successor as head of the Byrnes family, but decides to pass the role to Greg, naming him "The Godfocker." Despite Greg reluctantly accepting the role, Jack begins to suspect Greg of infidelity when he sees him with a drug representative, Andi Garcia (Alba), who openly flirts with him, and the presence of Sustengo erection pills in Greg's house, which prompts Jack to think Greg is no longer sexually attracted to his wife, Pam (Polo). After breaking up for the summer, the four friends regroup for a party held by Stifler (Seann William Scott), their overtly sex-crazed class clown and friend. His party is shut down thanks in part to the neighbors calling the police. Kevin (Thomas Ian Nicholas) is inspired by his brother to make his summer with his friends count by renting a beach home. He, Jim, Oz and Finch plan to take off to a beach house together, where they intend to spend the summer. Kevin realizes a problem and worked out that they can only afford the house if they have a fifth member to work along with them. Four high school seniors from western Michigan are good friends: Jim Levenstein (Jason Biggs), an awkward, nerdy and sexually naïve character whose dad Noah (Eugene Levy) attempts to offer sexual advice including purchasing and giving him pornography; Chris "Oz" Ostreicher (Chris Klein), a member of the high school lacrosse team; Kevin Myers (Thomas Ian Nicholas), their geeky leader who wants to lose his virginity with his girlfriend named Vicky (Tara Reid); and Paul Finch (Eddie Kaye Thomas), a mochaccino-drinking sophisticate and a nerd.
