Quelque part en Inde, au milieu d'un lac, un jeune couple se demande mutuellement en mariage sur une barque isolée. Une vache tombe soudainement du ciel et envoie la barque au fond de l'eau... Dans le Nord de la France, Pierre est un célibataire endurci, maniaque et bourru qui travaille dans une quincaillerie. Il passe ses journées à recompter le nombre de vis dans les boîtes de son fournisseur, et occupe ses soirées à collectionner les articles de faits divers insolites et morbides. Un jour alors qu'il se balade sur les quais déserts du port, il est accosté par un Indien violemment expulsé d'une camionnette. Totalement démuni et ne parlant que le bengali, celui-ci montre à Pierre un bout de carton où est inscrite une adresse située à l'autre bout de la ville. Pierre décide finalement de l'y emmener.

J. B. Bernstein (Jon Hamm) is a big time sports agent who, along with his partner Ash Vasudevan (Aasif Mandvi) recently formed their own company. Unfortunately, all of J.B.'s clients have retired, and he is unable to reel in star football player Popo Vanuatu (Rey Maualuga). Desperate to find new clients, J.B. realizes India, with over one billion people, has real potential for untapped baseball talent. He approaches investor Mr. Chang with his proposal - a talent contest staged in India called "Million Dollar Arm." Contestants score points by demonstrating they can pitch a baseball with speed and accuracy. Along with the prize money, two winners will be flown to the U.S. and receive coaching to become legitimate baseball prospects within two years. Chang commits to providing the funding, on the condition the prospects are ready within only one year. With no alternative, J.B. reluctantly assures Chang the winners will be ready for a major league try-out within one year.

The film begins where its predecessor ended, with reigning don Shoaib Khan (Akshay Kumar) earning respect after assassinating his mentor, Sultan Mirza (Ajay Devgn). Hardik (Kunal Khemu) and Luv (Vir Das) are two dope-heads, living with their roommate/friend Bunny (Anand Tiwari). After Hardik loses his job and Luv gets dumped by his girlfriend, both decide to tag along with Bunny to Goa to relax. In Goa, Luv falls for Luna (Puja Gupta), and she invites the guys to a rave party, organised by the Russian mafia, which is being held on a secluded island. At the party a new drug, D2RF, is launched. The next morning, the three roommates find that the island has been infected by the "living dead" (zombies), and the three men save luna from her villa and stay together in hopes of surviving. A Russian mafioso, Boris (Saif Ali Khan, originally from Delhi/Dilli), arrives and reveals himself as a zombie slayer. The three roommates and Luna plan to use the boat they had used to travel to the island to escape from it. Finally, after escaping the island, they discover that the mainland has also been infested by the zombies, forcing them to find another way to survive.

Maanav (Arunoday Singh) is a struggling filmmaker who will not compromise on the script he has written. His girlfriend Ruchi (Shahana Goswami), a successful film editor, arranges for him to meet film producer Nitin (Sushant Singh). Nitin likes the script but is not very sure of its box office potential. Maanav then suggests a story from the Panchtantra: A woman is caught red-handed with her lover by her husband and yet, she manages to wriggle out of it Scot-free! Nitin loves the story but finds it too short for a feature film. Maanav then creates three more stories based on the same premise: in a way, the Panchantra story travels in different versions to the modern times through the film. The four stories are woven together by a common story. Mirch itself echoes this structure, with four stories mingling with the main narrative.

Farhan Qureshi (R.Madhavan) and Raju Rastogi (Sharman Joshi) are students at the Imperial College of Engineering (ICE), a highly regarded Indian engineering college. Farhan's passion is wildlife photography, but he pursues an engineering degree to appease his father. Raju chooses engineering with hopes of improving his family's financial situation, but his lack of self-confidence results in poor grades. Their third roommate and friend, Ranchodas 'Rancho' Shamaldas Chanchad (Aamir Khan), has immense interest in robotics, and invents in his spare time.
