Tommy recently escaped from jail after being accused of murdering his manager, and asks the duo to accompany him on a visit to his fiancée, Helen Gray (Nancy Guild). He wants her uncle, Dr. Philip Gray (Gavin Muir), to inject him with a special serum he has developed which will render Tommy invisible, and hopes to use the newfound invisibility to investigate his manager's murder and prove his innocence. Dr. Gray adamantly refuses, arguing that the serum is still unstable, recalling that the formula's discoverer John Griffin was driven insane by the formula and did not become visible again until after he was killed. However, as the police arrive Tommy injects himself with it and successfully becomes invisible. Detective Roberts (William Frawley) questions Dr. Gray and Helen while Bud and Lou search for Tommy.

Dr. Luther Brooks (Sidney Poitier), an intern who has just passed the state board examination to qualify for his license to practice, is the first African-American doctor at the urban county hospital at which he trained. Because he lacks self-confidence, Luther requests to work as a junior resident at the hospital for another year. Johnny (Dick Paxton) and Ray Biddle (Richard Widmark), brothers who were both shot in the leg by a policeman as they attempted a robbery, are brought to the hospital's prison ward. As Luther tends to the disoriented Johnny, he is bombarded with racist slurs by Ray, who grew up in Beaver Canal, the white working class section of the city. Believing that Johnny has a brain tumor, Luther administers a spinal tap, but Johnny dies during the procedure. Wondering if Ray's antagonism may have caused him to be careless, Luther consults his mentor, chief medical resident Dr. Daniel Wharton (Stephen McNally), and Wharton concedes that a brain tumor was only one possibility. Feeling that he must prove the accuracy of his diagnosis, Luther requests an autopsy, but Wharton informs him that according to state law, they cannot proceed without the permission of the deceased's family. When Ray refuses, as he does not want his brother's body to be cut up, Wharton confers with the head of the hospital, Dr. Sam Moreland (Stanley Ridges), about requisitioning an autopsy.

The fabled city of Dzamm in Africa is rumoured to be a land of wealth. Looking to exploit its rich resources, Captain Rawlings (Ralph "Eddie" Dunn) dispatches two of his henchmen (Wally West and George DeNomand) to find the hidden trail to the city. After killing a few natives of Dzamm along the way, the inept servants of Rawlings are eaten alive by a large pride of wild lions. Nearby, jungle roamer Jungle Jim (Johnny Weissmuller) receives Li Wanna, the daughter of Dzamm's chief religious official Zoron (Nelson Leigh). Wanna seeks Jungle Jim's help in fighting the mercenaries led by Rawlings. It is revealed that the rumour is indeed true—stashed in the temple of Dzamm are crates of precious stones. After hearing news of the murdered Dzamm natives, Jungle Jim and Wanna hurry to the scene, just to find the henchmen's corpses. Seeing the lions, the agile adventurer and Wanna climb up a tree to safety. An African elephant approaches, scaring the beastly cats away. Jungle Jim and Wanna return to Zoron's residence, where they strategically discuss a peaceful resolution to get rid of Rawlings and his team. They come to the conclusion that giving Rawlings a few diamonds is the best idea. Meanwhile, Wanna's brother Chot (Paul Marion) is hypnotised by businessman Calhoun's (Joseph Vitale) niece Norina (Myrna Dell). Chot falls in love with Norina and unknowingly spills some secrets of Dzamm to her. Norina and Calhoun turn out to be under Rawling's payroll.
