In 1959, Alfred Hitchcock opens his latest film, North by Northwest, to considerable success, but is troubled by a reporter's insinuation that it is time to retire. Seeking to reclaim the artistic daring of his youth, Hitchcock turns down film proposals like adapting Casino Royale in favor of a horror novel called Psycho by Robert Bloch, which is based on the crimes of murderer Ed Gein. Gein appears in sequences throughout the film in which he seems to prompt Hitchcock's imagination regarding the Psycho story, or act as some function of Hitchcock's subconscious mind (for instance, drawing Hitchcock's attention to sand on his bathroom floor, the quantity of which reveals how much time his wife Alma has been spending at the beachhouse with Whitfield Cook).

Julian Marsh (Jake Hoffman), an unemployed young lothario, is forced by his doctor dad (Chip Zien) to accept a job directing an off-Broadway play called Rosencrantz and Guildenstern Are Undead, which is described as a weird adaptation of Hamlet. The play has been written by a mysterious, pallid Romanian named Theo Horace (John Ventimiglia), a vampire who has just killed a young woman (Bijou Phillips). Unaware of the danger that surrounds Theo’s play, Julian casts his best friend Vince (Kris Lemche) as Hamlet and uses his new job to impress his ex-girlfriend, Anna (Devon Aoki), an aspiring actress. To Julian’s dismay, Anna has taken up with a shady businessman named Bobby Bianchi (Ralph Macchio), who has reputed ties to the Mafia. When Anna is cast as Ophelia, she slowly gets involved with Theo. The roles of Rosencrantz and Guildenstern in Theo’s play are filled by Carlo (Carlos Velazquez) and Mickey (Mike Landry), two narcissistic actors as foolish as the characters they are cast to play. Opening night draws near and people begin to die. Two bumbling New York City detectives (Jeremy Sisto and Lou Carbonneau) chase suspects as the movie builds to its climactic sequence.

The film is narrated in flashback by Jake Briggs (Eric Stoltz), a young aspiring playwright, culminating in the production of one of his plays off-Broadway by agent Carl Fisher (Tony Curtis). The play is a flop, at least in part because the lead parts are given to two actors, Dana Coles and Jason Brett (Kathleen Turner and Chris Noth), who are "not right" for the roles. Along the journey, Jake reviews his relationships with girlfriend Joanne (Mary-Louise Parker), best friend Chris (Ralph Macchio), his mother Shirley (Jill Clayburgh), and his mostly absentee father Roman (Paul Guilfoyle).
