An opening prologue set years before the events of the first film tells that Lord Shen, the scion of a peacock clan that rules Gongmen City in ancient China, seeks to harness fireworks as a weapon. A dust speck is dislodged from its obscure place and sent adrift through the Jungle of Nool. At the same time, Horton the elephant (Jim Carrey), the jungle's eccentric nature teacher, takes a dip in the pool. The dust speck floats past him in the air, and he hears a tiny yelp coming from it. Believing that an entire society of microscopic creatures are living on that speck, he places it atop a clover. Horton finds out the speck harbors the city of Whoville and its inhabitants, the Whos, led by Mayor Ned McDodd (Steve Carell). He has a wife, Sally (Amy Poehler), 96 daughters (whose names all begin with the letter H), and one teenage son named JoJo (Jesse McCartney). Despite being the oldest and next in line for the mayoral position, JoJo doesn't want to be mayor, and because he's so scared of disappointing his father, never speaks. Once Horton begins carrying the speck with him, the city starts experiencing strange phenomena (earthquakes and changes in the weather), and the Mayor finds his attempts to caution Whoville challenged by the Town Council, led by the opportunistic yet condescending Chairman (Dan Fogler).

Alison Scott (Katherine Heigl) is a career-minded woman who has just been given an on-air role with E! and is living in the pool house with her sister Debbie's (Leslie Mann) family. He lives off funds received in compensation for an injury and sporadically works on a celebrity porn website with his roommates (Jay Baruchel, Jonah Hill, Jason Segel and Martin Starr), in between smoking marijuana or going off with them at theme parks such as Knott's Berry Farm. While celebrating her promotion, Alison meets Ben at a local nightclub. After a night of drinking, they end up having sex. Due to a misunderstanding, they don't use protection: Alison uses the phrase "Just do it already" to encourage Ben to put the condom on, but he misinterprets this to mean to dispense with using one. The following morning, they quickly learn over breakfast that they have little in common and go their separate ways, which leaves Ben visibly upset.

For years, the Republic of Wadiya (located in the Horn of Africa, supplanting the actual nation of Eritrea) has been ruled by Admiral General Haffaz Aladeen (Sacha Baron Cohen), a childish, tyrannical, sexist, anti-western, and antisemitic despot who surrounds himself with female bodyguards, sponsors al-Qaeda (specially giving shelter to Osama Bin Laden after "they killed his double one year ago") and is working on developing nuclear weapons to attack Israel. The film is divided into three acts set in the lead-up to key product launches hosted by Apple co-founder Steve Jobs (Fassbender). The first depicts Jobs and marketing executive Joanna Hoffman (Winslet) dealing with problems before the Apple Macintosh launch in 1984. The computer's voice demo will not say "hello", and Jobs demands engineer Andy Hertzfeld (Stuhlbarg) fixes it, threatening to publicly humiliate him in the presentation. Hertzfeld finally suggests faking the demo using the prototype 512k computer. Meanwhile, Jobs rants about a Time magazine article that exposed his paternity dispute with ex-girlfriend Chrisann Brennan (Waterson): Jobs denies he is the father of Brennan's five-year-old daughter, Lisa. Brennan arrives with Lisa to confront Jobs; she is bitter over his denials and his refusal to support her despite his wealth. Jobs bonds with Lisa over her MacPaint art and agrees to provide more money and a house, though he still denies his fatherhood. Apple co-founder Steve Wozniak (Rogen) wishes Jobs well and asks him to acknowledge the Apple II team in his remarks, but Jobs refuses. Flashbacks show the two creating the Apple II, revealing that Wozniak's ideas made the product successful. Jobs then discusses company politics with CEO John Sculley (Daniels); they talk about Jobs' life as an adopted child, and Jobs admits that his need for control stems from his feelings of powerlessness in being given up.
