Tyler Perry Presents Peeples centers around the Peeples, an affluent East Coast family celebrating their annual reunion for Moby Dick Day – a Peeples family tradition at Sag Harbor in the Hamptons. Their weekend is interrupted when Wade Walker (Robinson), the fiance of their daughter Grace (Washington) crashes to propose marriage after being goaded by his brother Chris. Upon arrival he loses his wallet while being chased by the family dog. While at the grocery store Wade notices Virgil (David Allen Grier), Grace's father and a judge having a conversation with an unidentified woman, who is later revealed to be the mayor of the town, as well as a conversation between Gloria (Hawk), Grace's Sister and her lover Meg;(Lewis-Davis) unknown to the family. As the time goes on, Wade begins to learn other things about Grace, such as her older ex-boyfriends, and the fact that she has breast implants. Wade also discovers that Grace's mother Daphne (S. Epatha Merkerson), was a professional singer during the 70's, as well as a recovering alcoholic.

The film centers on two main characters: Lazarus Redd (Samuel L. Jackson), a deeply religious farmer and former blues guitarist, and Rae Doole (Christina Ricci), a young sex addict. The Great Gonzo has always been identified as a "whatever". But after he begins having disturbing dreams of abandonment and rejection, he begins to realize just how alone he is in the world. One of his nightmares involves him being denied entry onto Noah's Ark by Noah (F. Murray Abraham). The next morning, Gonzo tells Kermit the Frog that he is getting tired of being called a "whatever." After an alien race appears to be trying to send him a message through bowls of cereal, Gonzo realizes that he may not be so alone after all and climbs to the rooftop to start watching the sky. Using a bolt of lightning, Gonzo communicates with a pair of cosmic fish, revealing to him that he is an alien from outer space.

Elmo is playing with his blanket in his house. After he bumps his blanket on a juice cup and after drying off his blanket at the laundromat, Elmo sees Zoe feeling depressed because her father cannot take her to the zoo, so he decides to make Zoe happy again by imitating certain zoo animals (a lion, a monkey, and a pig). When Zoe sees his blanket, Elmo refuses to share, resulting in a tug-of-war that rips Elmo's blanket. Elmo is furious and declares that Zoe is no longer his friend. Suddenly, Telly Monster inadvertently takes away the blanket while rollerskating out of control. They go around Finders Keepers and knock over a tray of drinks Ruthie is carrying. Telly hits Cookie Monster at the revolving door of Furry Arms hotel. The blanket accidentally lands in the hands of Oscar the Grouch, who drops it in his trash can after sneezing on it. Elmo drops into the bottom of Oscar's trash can, where he finds the blanket nailed to a door. But they are both teleported to Grouchland USA, a world filled with Grouches where a greedy man named Huxley (Mandy Patinkin) steals anything he can grab including Elmo's blanket. Elmo is determined to find his blanket and begins a journey through Grouchland. He asks a kind girl named Grizzy to help in his quest, but she later abandons him when Elmo discovers that Huxley's house is on the top of the faraway Mount Pickanose.

Millionaire animal lover and overall spoiled rich girl Trudy (Rene Russo) adds Buddy, a gorilla, to her family. Buddy finds life in the city very difficult to deal with. Although Trudy raises him as her own son in her mansion (which also houses a few comical chimpanzees, a kitten, a horse, a cheeky, talking green parrot, her prize-winning champion pack of Briards, a raccoon and a flock of geese), he becomes hard to control due to his strength. A particularly bad experience in the Chicago World's Fair makes things even harder for Buddy. After he goes on an aggressive rampage and nearly destroys Trudy's home, Buddy is taken to an ape sanctuary to live among his own kind in peace.
