Kamal Mehra (Anil Kapoor) is a businessman who owns Ayka, a company on the verge of bankruptcy. He has a troubled relationship with his wife, Neelam (Shefali Shah), and two children, Ayesha (Priyanka Chopra) and Kabir (Ranveer Singh). Ayesha owns a successful travel business of her own but is unhappy with her marriage to Manav (Rahul Bose), who is controlling, narcissistic and disdains Ayesha's family. Kabir unwillingly participates in the family business even though he yearns to become a pilot. In celebration of Kamal and Neelam's 30th wedding anniversary, they invite their family and friends for a 10-day cruise across the Mediterranean. Among their invitees are Mr. Sood (Parmeet Sethi) and his family. Kamal plots to get Kabir married to Sood's daughter, Noorie (Riddhima Sud), in exchange for his investing in Ayka.

Lootera is inspired by O. Henry's The Last Leaf. In the picturesque town of Manikpur, West Bengal in 1953, a landlord goes to watch a Chhau dance with his daughter, Pakhi Roy Chaudhary (Sonakshi Sinha), an aspiring writer. After Pakhi has an asthma attack from the ensuing excitement, she is rushed home and given medication.
