L'histoire se déroule cinq ans après les événements de Sharknado 3. La tempête porteuse de requins tueurs est de retour et la cible des squales est à présent Las Vegas. Wolf (Patrick Warburton), Granny (Glenn Close), and Twitchy (Cory Edwards) are on a rescue mission to save Hansel and Gretel (Bill Hader and Amy Poehler) from an evil witch named Verushka (Joan Cusack). The plan goes wrong however and Granny is kidnapped as well. To celebrate his upcoming marriage to Tracy Garner (Sasha Barrese), Doug Billings (Justin Bartha) travels to Las Vegas with his best friends Phil Wenneck (Bradley Cooper) and Stu Price (Ed Helms), as well as Tracy's brother and Doug's future brother-in-law Alan (Zach Galifianakis), in Doug's future father-in-law's vintage Mercedes-Benz. They stay at Caesars Palace, where they relax in the room before celebrating with a few drinks on the hotel rooftop. The next morning, Phil, Stu, and Alan awaken to find they have no memory of the previous night, and Doug is nowhere to be found. Stu is missing a tooth, their hotel suite is in disarray, a tiger is in their bathroom, a chicken in their living room, and a baby is in the closet, whom they name "Carlos". They find Doug's mattress impaled on a statue outside of their hotel and when they ask for their Mercedes, the valet delivers an LVPD police cruiser.

Following release from prison, Daniel "Danny" Ocean (George Clooney) violates his parole by traveling to California to meet his partner-in-crime and friend Rusty Ryan (Brad Pitt) to propose a caper. The two go to Las Vegas to pitch the plan to wealthy friend and former casino owner Reuben Tishkoff (Elliott Gould). The plan consists of simultaneously robbing the Bellagio, The Mirage, and the MGM Grand casinos. Reuben's familiarity with casino security makes him very reluctant to get involved, but when he starts to think of it as a good way to get back at his rival, Terry Benedict (Andy García), who owns all three casinos, Reuben agrees to finance the operation. Jerry Logan, a cab driver in Las Vegas, picks up a nervous passenger who offers him $100 to get to the airport as quickly as possible. After returning home from South Korea to great acclaim, three members of the U.S. National Karate Team (Tommy Lee, Alex Grady, and Travis Brickley) set up their own martial arts studio in Las Vegas. Unbeknownst to his friends, however, Travis has been competing at "The Coliseum", a brutal underground fighting arena run by a shady promoter named Weldon (Wayne Newton), whose protégé Brakus (Ralf Möller) is the owner and undefeated champion. Though the rules of the Coliseum state that a challenger must defeat three of its "Gladiators" in order to face Brakus, Travis wastes little time in challenging Brakus outright.
