In 1890, all of New York City is excited because widowed, brassy Dolly Levi is in town ("Call On Dolly"). Dolly makes a living through matchmaking and numerous sidelines ("Just Leave Everything to Me"). She is currently seeking a wife for grumpy Horace Vandergelder, the well-known "half-a-millionaire", but it becomes clear that Dolly intends to marry Horace herself. Dolly travels to Yonkers, New York to visit Horace. Ambrose Kemper, a young artist, wants to marry Horace's weepy niece, Ermengarde, but Horace opposes this because Ambrose's vocation does not guarantee a steady living. Horace, who is the owner of Vandergelder's Hay and Feed, explains to his two clerks, Cornelius Hackl and Barnaby Tucker, that he is going to get married because "It Takes a Woman" to cheerfully do all the household chores. He plans to travel to New York City to propose to Irene Molloy, who owns a hat shop there. Dolly arrives in Yonkers and sends Horace ahead to the city. Before leaving he tells Cornelius and Barnaby to mind the store.

A workhouse in Dunstable, England is visited by the wealthy governors who fund it. While a sumptuous banquet is held for them, the barefoot orphan boys who work there are being served their daily gruel. They dream of enjoying the same "Food, Glorious Food" as their masters. While eating, some boys draw straws to see who will ask for more to eat, and the job falls to a boy named Oliver Twist. He goes up to Bumble and Widow Corney, who run the workhouse and serve the gruel, and asks for more. Enraged, Bumble takes Oliver to the governors to see what to do with him ("Oliver!"). A decision is made to have Oliver sold into service. Bumble parades Oliver through the snow, trying to sell him to the highest bidder ("Boy for Sale"). Oliver is sold to an undertaker named Mr. Sowerberry, who intends to use him as a mourner for children's funerals. After his first funeral, Noah Claypole, Sowerberry's apprentice, insults Oliver's mother. Oliver attacks Noah in fury and Sowerberry forces him into a coffin while Noah fetches Bumble. Oliver is too angry to be intimidated by Bumble, who places the blame on not keeping Oliver on a diet of gruel, instead of meat, which made him strong. Oliver is thrown into the cellar as further punishment. Alone in the dark with a roomful of empty coffins, Oliver wonders ("Where is Love?"). While clutching the window grate, Oliver pushes it open and escapes.

In the city of Rome during the reign of Emperor Nero, Pseudolus (Zero Mostel) is "the lyingest, cheatingest, sloppiest slave in all of Rome", whose only wish is to buy his freedom from his master's parents, the henpecked Senex (Michael Hordern) and his dominating wife Domina (Patricia Jessel). When he finds out that his master, Senex's handsome but dim son Hero (Michael Crawford), has fallen in love with Philia (Annette Andre), a beautiful virgin courtesan from the house of Marcus Lycus (Phil Silvers), buyer and seller of beautiful women next door, Pseudolus makes a deal: he will get the girl for Hero in return for his freedom. Unfortunately, the virgin has been sold to the great Roman soldier, Captain Miles Gloriosus (Leon Greene), who even now is on his way from conquering Crete to claim her as his bride. Pseudolus blackmails his overseer, Hysterium (Jack Gilford) into masquerading as the corpse of Philia to fool the captain, but things go wrong at every turn.

Maria is a free-spirited young Austrian woman studying to become a nun at Nonnberg Abbey in Salzburg in 1938. The Mother Abbess, believing Maria would be happier outside the abbey, sends her to the villa of retired naval officer Captain Georg von Trapp to be governess to his seven children—Liesl, Friedrich, Louisa, Kurt, Brigitta, Marta, and Gretl. The Captain has been raising his children alone using strict military discipline following the death of his first wife. At first, the children treat Maria as they did their former governesses—playing tricks on her as a way of gaining their father's attention. In Edwardian London, Professor Henry Higgins (Rex Harrison), a scholar of phonetics, believes that the accent and tone of one's voice determines a person's prospects in society. In Covent Garden one evening he boasts to a new acquaintance, Colonel Hugh Pickering (Wilfrid Hyde-White), himself an expert in phonetics, that he could teach any person to speak in a way that he could pass them off as a duke or duchess at an embassy ball. Higgins selects as an example a young flower seller, Eliza Doolittle (Audrey Hepburn), who has a strong Cockney accent. Eliza's ambition is to work in a flower shop, but her thick accent makes her unsuitable. Having come from India to meet Higgins, Pickering is invited to stay with the professor. The following morning, Eliza shows up at Higgins' home, seeking lessons. Pickering is intrigued and offers to cover all expenses if the experiment should be successful.

Masquerading as a traveling band instructor, Professor Hill plans to con the citizens of River City into paying him to create a boys' marching band, including instruments, uniforms, and music instruction. Once he has collected the money and the instruments and uniforms have arrived, he will hop the next train out of town, leaving them without their money or a band.
