The film begins with the recovery of Simar (Priyanka Chopra) and her husband Nikhil (Arjun Rampal) from a car accident. Simar is almost unscratched but Nikhil has amnesia, along with major physical damage. Simar is determined that Nikhil will get better and resume his life, and brings in the best surgeons to reconstruct his damaged leg and face, and then later helping him with his physical therapy.

Two childhood friends grow up to be on the opposite sides of law, one an Additional Police Commissioner, Mahendra Ashwini Kumar, and the other a crime lord, Raj Pratap Singhal. An associate of Singhal kills the son of Mahendra, and his other son, Rajesh, also a police officer, swears to avenge the death. Mahendra's deceased son leaves behind a widow, Kaushalya and a daughter, Meghna. On a chance meeting, Meghna meets with Vicky Malhotra, and eventually both in love with each other. The Kumars are introduced to Vicky, and they approve of him. Mahendra is then promoted as the Commissioner of Police, and is subsequently killed by another associate of Singhal. With the police closing in on Singhal, he decides to re-locate to Dubai, U.A.E. with his wife, son, daughter-in-law Anu, and he arranges the abduction of Meghna to ensure his safety from Rajesh's wrath. It is then that Meghna comes to know that Vicky is really the other son of Singhal, and who now has started showing his true colors.

Lalita (Madhuri Dixit) catches the eye of JB (Raj Babbar) and he immediately falls in love with her and decides that he must have her. Her uncle Madhi arranges for them to marry but he is only doing this because JB is paying him to. When she refuses, JB kidnaps her and keeps her in his home until the day of the wedding. She is literally watched over by JB every day in case she tries to escape. On the day of the wedding, Lalita faints and uses this to escape. She also moves her mother away and barely escapes JB who assumes that she has been killed after he saw her being hit by a car. In fact, Lalita swapped her wedding dress with someone else, so the person that JB saw being hit by the car was not Lalita.

Naina (Deepika Padukone) is a shy and nerdy medical student. She studies constantly and always tops her classes, but she feels like an outcast. Dushant (played by Khan), Jaya (Tandon), Sujit (Behl) and Nisha (Nishan) are childhood friends. Dushant aspires to become a successful singer but has little money. Jaya helps him to realize his ambitions. As Dushant achieves success, a rift is created between the two of them. Sujit and Nisha take undue advantage of this to get close to Dushant and create a misunderstanding between him and Jaya. Kamlesh Dhingra (Avtaar Gill), Nisha's father, approaches Dushant for financial assistance for his business venture but is turned down by Rajpal (Kiran Kumar), Dushant's mentor. Dhingra and Sujit plot to amass Dushant's wealth by luring Dushant to marry Nisha and getting rid of Rajpal. Dushant learns of this plot. When Sujit is murdered, Rahul (Amit Kumar), a police officer, comes to investigate the murder. Nisha is also murdered under mysterious circumstances and the climax builds when Dhingra is also killed.

Lallan Singh (Abhishek Bachchan) is a goon, originally from Bihar but settled in Kolkata, West Bengal because his brother Gopal Singh (Sonu Sood) has left him alone and he had no option of earning back home. Michael is in love with his neighbour Radhika (Esha Deol) who lives with her uncle and aunt. And then there is Arjun (Vivek Oberoi), carefree and spoiled son of an IAS officer. He wants to relocate to the U.S. for a better future. He falls in love with Mira (Kareena Kapoor), whom he just met. One day, Arjun and Mira get into a quarrel on the road and Mira gets into an auto. Arjun gets lift from Michael to catch up with Mira, who is going that way. Suddenly, Michael is hit by a bullet and he is saved by Arjun. After talking to Michael, Arjun changes his mind and wants to be a politician. Prosenjit is worried when he hears news of students standing in the election. He uses every possible way to get them out of Politics.Firstly he provides scholarship of a prestigious foreign university to Michael. When Michael refuses the bribe, he orders his goon Gopal Singh to take control. Gopal beats some students but faces very strong retaliation from Michael and his fellow students. After that Lallan Singh takes charge and kills Gopal. He kidnaps Arjun and other candidates. However, they manage to escape with the help of Lallan Singh's ally. Lallan follows Arjun and beats him up. While running, Arjun manages to call Michael for help. He comes to rescue Arjun at Howrah Bridge.

Zubeidaa acts in films secretly, but when her father finds out he forbids her to carry on and quickly arranges her marriage to Mehboob Alam (Vinod Sharawat). Things seem happy for her when she gives birth to Riyaz. However, a disagreement arises between Suleman and Mehboob's father, and Mehboob divorces Zubeidaa few days after she gives birth.
