The film starts in a courtroom where a Brahmin woman Janaki (Meena) is fighting for divorce from her husband Pandian (Kamal Haasan). She offers various reasons for wanting a divorce. She has married Pandian against the wishes of her rich father Vishwanathan Iyer (Gemini Ganesan). But she isn't able to come to terms with living in Pandian's small house without amenities. Pandian is an assistant dance director with a modest income and is not able to spend much time with the family. All this accumulates and Janaki decides to apply for divorce, which is granted. The court orders that Pandian can meet his daughter Barathi, once a week and this is his only solace. His daughter, however, loves him a lot and doesn't like the arrangement.

The story, based on an idea from Salman Khan, centres on Saajan, the son of a colonel in the Indian army, and Kaajal, a modest girl from "a respectable family." The film opens with Saajan travelling in a bus, when he catches a glimpse of Kaajal on another bus, and they are both smitten. They do not formally meet and, since Saajan is off to start at college, he does not think he will ever see her again. But his new friends at college, Buddha, Tempo and Refill, one night insist on visiting a brothel in a seedier part of Bombay. Saajan only reluctantly agrees, but ultimately refuses to select a prostitute — until he hears a new girl being beaten by her pimp and decides to protect her. To his surprise, it is Kaajal (called Paro at the brothel), who has been kidnapped by a pimp after she was tricked by a job offer in Bombay.

The film begins in an illegal underground dog-fighting arena in Korea, where an FBI agent named John Maxwell (Paul Giamatti) has been identified. John is ordered to be killed by a Korean mob boss, but is eventually rescued by his undercover partner and master of disguise, Malcolm Turner (Martin Lawrence). Suddenly, a group of FBI agents storm around the arena. Meanwhile, a criminal named Lester Vesco (Terrence Howard), who was originally serving a life sentence in prison for murder and armed robbery, escapes from his cell by killing the doctor and stealing his car. The FBI assigns Malcolm and John to capture Lester by sending them to small-town Cartersville, Georgia to stake out the house of a fat, elderly African American woman named Hattie Mae Pierce (Ella Mitchell; whom her friends call "Big Momma"), the estranged Southern grandmother of Lester's ex-girlfriend, Sherry Pierce (Nia Long), who supposedly aided Lester in his original bank robbery by giving him the key to the vault. After Big Momma unexpectedly leaves town to help her ill friend within a couple of weeks, Malcolm and John sneak into her house to plant security cameras and tap the phones. Sherry later calls Big Momma's house and Malcolm disguises his voice as Big Momma in order to lure Sherry and possibly obtain a confession. The plan works, in which Malcolm and John work together on a Big Momma disguise costume before Sherry's arrival the next day.

Malcolm, in an attempt to capture Russian gang member Chirkoff (Tony Curran), uses an informant named Canetti to deliver a flash drive to the gang, while Trent attempts to ambush Malcolm on the job. Canetti (Max Casella) reveals that the flash drive is empty and a duplicate is hidden with a friend at the Georgia Girls School for the Arts.

Malcolm Turner (Martin Lawrence) has been assigned a desk job in public relations as an FBI agent, since he wants to live with his new wife, Sherry Pierce (Nia Long), during her delivery for the couple's new baby boy. Meanwhile, an incident occurs in Orange County, California, where Malcolm's old friend, Doug Hudson (Kirk B.R. Woller), has been killed while he was going undercover. FBI agent Kevin Keneally (Zachary Levi) is doing surveillance on a former U.S. Army military intelligence specialist named Tom Fuller (Mark Moses), who has since retired and is working for a private corporation called National Agenda Software. The FBI has soon discovered that Tom is developing a computer worm which will create backdoors into the databases of all the branches of the U.S. government. Affected by his friend's death, Malcolm asks FBI chief, Crawford (Dan Lauria), to put him on the case, but Crawford refuses and tells him to stay away for safety analysis. By eavesdropping via the webcam, Malcolm finds out that the FBI is sending one of the agents to infiltrate Fuller's house as a nanny. Giving Sherry the pretext of attending a safety conference in Phoenix, Arizona, Malcolm leaves for Orange County and takes the "Big Momma" costume with him.

Ĥamida (Suhair Ramzi) is the daughter of an illiterate widower, al-Ĥag Firghalee. Firghalee attempts to prevent his daughter from studying at a university, so the medical student Ĥassan helps her. After Firghalee reveals he will marry Ĥamida to a boy she dislikes, Ĥassan has her disguise herself as a man, Mahmoud. Ĥassan and his friends lie to Firghalee, telling him they performed transsexual surgery to change Ĥamida into a man. Ĥamida's disguise has an effeminate appearance, and both male and female employees at Firghalee's furniture shop are attracted to "Mahmoud" and many conclude "he" is a homosexual.

In the 1970s, Black Dynamite, a Vietnam War veteran and former CIA officer, vows to clean up the streets of drug dealers and gangsters after his younger brother Jimmy is killed by a shady organization. O'Leary, Black Dynamite's former army and CIA partner, reinstates him into the agency because they do not want him seeking vengeance by himself. While trying to get to the bottom of Jimmy's murder, he finds out that his brother was actually working undercover for the CIA. Black Dynamite also discovers the shady organization is filling the black orphanages with heroin. He declares war on local drug dealers and successfully cleans up the streets, earning him the affection of Gloria, a black power activist who works at the local orphanage.
