In preparation for the latest broadcast of the popular quiz show Twenty One, the questions and answers to be used are transported from a secure bank vault to the studio as producers Dan Enright (David Paymer) and Albert Freedman (Hank Azaria) watch from the control booth. The evening's main attraction is Queens resident Herb Stempel (John Turturro), the reigning champion, who correctly answers question after question. However, both the network, NBC, and the corporate sponsor of the program, a supplementary tonic called Geritol, find that Stempel's approval ratings are beginning to level out, meaning the show would benefit from new talent.

The film is narrated in flashback by Jake Briggs (Eric Stoltz), a young aspiring playwright, culminating in the production of one of his plays off-Broadway by agent Carl Fisher (Tony Curtis). The play is a flop, at least in part because the lead parts are given to two actors, Dana Coles and Jason Brett (Kathleen Turner and Chris Noth), who are "not right" for the roles. Along the journey, Jake reviews his relationships with girlfriend Joanne (Mary-Louise Parker), best friend Chris (Ralph Macchio), his mother Shirley (Jill Clayburgh), and his mostly absentee father Roman (Paul Guilfoyle). The film ends with Jake and Joanne going their separate ways, mostly because of competing career goals, and Jake hoping to write more plays with greater success.

Danny Aiello stars as Harry Stone, an NYC film director who has been living in Paris, France for the past ten years. Two children, Jacques Mayol (Jean-Marc Barr) and Enzo Molinari (Jean Reno), have grown up on the Greek island of Amorgos in the 1960s. They challenge each other to collect a coin on the sea floor and Jacques loses. Later Jacques' father — who harvests shellfish from the seabed using a pump-supplied air hose and helmet — goes diving for shellfish. His breathing apparatus and rope gets caught and punctured by rocks on the reef and weighed down by water, he drowns. Jacques and Enzo can do nothing but watch in horror as he is killed.

Nikki Finn is a carefree young woman, who is always dressed in a leather jacket and skirt, with fire-red lips, platinum bob hair and speaking in a high-pitched voice. One day, her boyfriend Johnny uncovers two men stealing money out of a trust fund and takes pictures of the theft. Johnny puts the pictures in a safety deposit box and gives Nikki the key for safekeeping. The thieves catch Johnny and murder him, then frame Nikki by putting his body into the trunk of her car. Fictional television station WIDB-TV (channel 8) experiences problems with its late-night airing of science-fiction classic Amazon Women on the Moon, a 1950s B movie in which Queen Lara (Sybil Danning) and Captain Nelson (Steve Forrest) battle exploding volcanoes and man-eating spiders on the moon. Waiting for the film to resume, an unseen viewer begins channel surfing—simulated by bursts of white noise—through late night cable, with the various segments and sketches of the film representing the programming found on different channels. The viewer intermittently returns to channel 8, where Amazon Women continues to resume airing before faltering once more.

Paul Hackett, a word processor, meets Marcy Franklin in a local cafe in New York. They discuss their common interest in Henry Miller. Marcy leaves Paul her number and informs him that she lives with a sculptor named Kiki Bridges, who makes and sells plaster of Paris paperweights resembling cream cheese bagels. Later in the night, under the pretense of buying a paperweight, Paul visits Marcy, taking a cab to her apartment. On his way to visit Marcy, a $20 bill is blown out the window of the cab, leaving him with only some spare pocket change. The cab driver is furious that he cannot pay, thereby beginning the first in a long series of misadventures for Paul that turn hostile through no fault of his own. At the apartment Paul meets the sculptor Kiki and Marcy, and comes across a collection of photographs and medications which imply that Marcy is severely disfigured from burns on her legs and torso. As a result of this implication, and as a result of a strained conversation with Marcy, Paul abruptly slips out of the apartment.
