The film follows the life of a middle-aged housekeeper, Séraphine Louis, who has a remarkable talent for painting. Untaught and following what she regards as religious inspiration she finds great appreciation in the beauty found in nature, especially her daily walks to work where she proudly and humbly stops to gaze at trees. In the beginning, it is noted that she stops to collect soil from plants as well as some blood from a dead pig. Later, in her small home lit by candles she is seen using these same ingredients while creating her art. At one point when her art begins to be seen, she is asked how she achieves the unusual effect in her "rouge" (reds). She replies that she prefers to keep that a secret.

The film begins with the contorted body of the woman, covered in frost. From this image, an unseen and unheard interviewer (the voice of Varda herself) puts the camera on the last men to see her and the ones who found her. The action then flashes back to the woman, Mona (Sandrine Bonnaire) walking along the roadside, hiding from the police and trying to get a ride. Along her journey she meets and takes up with other vagabonds such as herself as well as a Tunisian vineyard worker, a family of goat farmers, a professor researching trees, and a maid who envies what she perceives to be a beautiful and passionate lifestyle. Mona explains to one of her temporary companions that at one time she had an office job in Paris and did very well for herself, but she became unsettled with the way she was living – choosing instead to wander the country free from any responsibility, picking up what she could to survive as she goes. Throughout the film, Mona's condition seems to become progressively worse until she finally falls where we first saw her, frozen and entrenched in her misery in a ditch.

Taking place in the Southern United States between Winter 1909 and Autumn 1937, the movie tells the life of a poor African American woman named Celie Harris (Whoopi Goldberg) whose abuse begins when she is young. By the time she is 14, she has already had two children by her father Alphonso "James" Harris (Leonard Jackson). He takes them away from her at childbirth and forces the young Celie (Desreta Jackson) to marry a wealthy young local widower Albert Johnson, known to her only as "Mister" (Danny Glover), who treats her like a slave. Albert makes her clean up his disorderly household and take care of his unruly children. Albert beats and rapes her often, intimidating Celie into submission and near silence. Celie's sister Nettie (Akosua Busia) comes to live with them, and there is a brief period of happiness as the sisters spend time together and Nettie begins to teach Celie how to read. This is short-lived; after Nettie refuses Albert's predatory affections once too often, he kicks her out. Before being run off by Albert, Nettie promises to write to Celie saying, "Nothing but death can keep me from it!".
