Bugs Bunny, looking for a place to pass the night, happens on an abandoned farm house, which, unbeknownst to Bugs, is the hideout of two gangsters, Rocky and Hugo. After claiming "Huh! Sounds like Inner Sanctum" while opening the squeaky front door, he drills a hole in the ground, dons a nightcap, descends in a manner as if walking down spiral stairs and goes to sleep. Shortly thereafter, Rocky and Hugo return pursued by rival gangsters (turning a corner where a billboard advertises Hotel Friz, an in-joke referring to director Friz Freleng). The running gunfight continues as they take cover inside the farmhouse; Bugs comically gets up in the middle of the gunfight (now also wearing a nightshirt) to use the bathroom and get a glass of water before returning to bed just as the shooting ends.

While traveling from Silver Creek, Colorado to his home in New York City, he is caught in a blizzard and his car goes off the road, rendering him unconscious. Paul is rescued by a nurse named Annie Wilkes (Kathy Bates), who brings him to her remote home. When Paul regains consciousness he finds himself bedridden, with both his legs broken as well as a dislocated shoulder. Annie claims she is his "number one fan" and talks a lot about him and his novels. As a reward for saving him, Paul gives Annie his new manuscript which she saved from the wreckage. While feeding him, she is angered and spills soup on him but regains control and apologizes. She buys a copy of Paul's most recently published book, Misery's Child, giving glowing praise to Paul as she progresses through the book. However, when Annie discovers that Misery dies at the end of the book she flies into a rage, almost smashing a table on Paul's head. She reveals that she lied about calling his agent and the authorities; nobody knows where he is. Annie leaves and Paul tries to escape from his room, but she has locked the door.

At the orphanage where she lives, the caretaker known as the Gray One tells scary stories about the evil nature of the bears that live in the outside world, though Celestine doubts they are entirely true. She loves to draw but must soon study dentistry, since that's what all rodents do, and to prepare, she must travel above ground to collect bear cubs' lost teeth from underneath pillows. On one such occasion, the cub's family catches Celestine in the act and chase her into a trash can where she is trapped and spends the night. The next morning, a destitute and starving bear named Ernest discovers Celestine and attempts to eat her. Celestine convinces him to let her go by helping him break into the basement of a candy shop, where he can eat his fill. He is soon caught by the store's owner, however, and arrested. Celestine, who is behind on her quota of collected teeth, agrees to free him from the police wagon if he will help her break into and rob the teeth from the office of the store owner's wife, who happens to be a dentist.

The movie opens with Mystery, Inc. being pursued by a moat monster. After an accident with Scooby-Doo (Scott Innes), he is caught and revealed to be a counterfeiter. This is actually an old case of the now-dissolved Mystery, Inc., who went their separate ways because they were frustrated with their mysteries having all involved mere crooks in monster costumes, as told on a talk show program by Daphne Blake (Mary Kay Bergman), who now, along with Fred Jones (Frank Welker), is running a successful TV series Coast to Coast with Daphne Blake, which involves her and Fred going to historic and inspiring places all across the country. For the show's upcoming second season, Daphne intends to visit haunted and mysterious places and track down real ghosts. When Daphne confesses on air that she misses the gang and that she wishes they were with her, Fred arranges a reunion and contacts Velma Dinkley (B.J. Ward), who has gone on and become the proprietor of a mystery bookstore, and Scooby and his owner Shaggy Rogers (Billy West), who have been bouncing from job to job, including working in customs at an airport, from which they were just fired after eating all the confiscated imported foods. The reunited gang surprise Daphne on her birthday and they go for a trip to Louisiana for her show.
