The film takes place in Australia and Vampire Rock, a rock formation shaped like a vampire head. There is a legend of a vampire named the Yowie Yahoo, who lives in the rock. The film starts at Vampire Rock where the "Vampire Rock Music Festival" is being set up. Many people are excited for it, but some don't think it should take place because it would anger the Yowie Yahoo. One such person is Malcolm Illiwara, even though his own grandson Daniel is the manager of the contest. One night, as Malcolm and his grandson watch a sure-to-win performer named Matt Marvelous, the Yowie Yahoo appears. The Yowie Yahoo and its three vampire minions capture Matt Marvelous and take him away. Everybody is scared, and Malcolm blames the contest for what happened.

The film begins with an introductory song by Tulie, a bayou dweller, who tells the story of Valdronya and the Mystery Inc. Gang. After solving the mystery of a Cockroach Monster that terrorized a factory, Fred, Daphne, Velma, Shaggy, and Scooby decide to take a much needed vacation. Velma drives everyone to Chauve Sourie Ville, a small town in Louisiana which is hosting a vampire festival called "Vampire Palooza." They meet Lita Rutland, the festival organizer, who explains that tourism is down and the festival is losing money. They also meet their host, Vincent Van Helsing, who runs a vampire museum and is an aspiring author, although his books are not selling. Jesper Poubelle, an anti-vampire agitator, is also stirring up trouble by protesting the festival. Van Helsing takes the Gang back to his museum and shows them various jewelry and a glass-faced casket containing the inert body of Valdronya, a centuries-old vampire.

The Joker and the Penguin break out of Arkham Asylum and race each other to find a stash of stolen money hidden within a crypt in Gotham Cemetery after an inmate, who had hidden it, told them about it. The Joker is quickly intercepted by the Batman and is electrocuted when he falls into a river by his super-charged joy buzzers while the Penguin takes the opportunity to enter the cemetery himself. While searching through a crypt, the Penguin accidentally cuts his hand while using his umbrella-sword to open a coffin he hopes contains the money, finding a body instead. Blood from his hand drips on the corpse's heart, which brings it to life; it is none other than the vampire lord Count Dracula, his body having been moved from Transylvania to Gotham City after his "death." After attacking and turning a watchman at the cemetery into a vampire, Dracula hypnotizes the Penguin into becoming his non-vampiric servant so that he may lead him through Gotham to feed and guard his coffin during the day while he regains his strength.
