Collier (Brown) enters prison, having been found guilty of killing her husband. She is introduced to the beautiful occupants of her cell, in for crimes ranging from political insurgency to heroin addiction. The women often clash, which leads to their torture by sadistic guard Lucian (Kathryn Loder). The torture ceremonies are viewed by an impassive cloaked figure.
Dorothy "Dot" Burton (Faye Emerson) is a member of a gang of bank robbers. Using her femininity and a cute dog provided her by her male cohorts who dognapped him, she is able to enter a bank before opening time, leaving the door open and the bank guard holding her dog, thus enabling a successful robbery. When police interfere with the getaway she faints and proclaims her innocence, however the police have strong doubts as "her" dog won't come to her and has a different name on his collar than what she calls him. After she confesses to her part in the robbery, she is sent to women's prison where she makes enemies of fellow inmates seeking Dot's share of the money.

As the night unfolds, an argument ensues in the car and Vince tells the others to get out, leaving Donna as his only passenger. When she asks him if he is worried about the other couple notifying the police, Vince tells her that he would murder anybody who ever turned on him. Subsequently, the police attempt to pull Vince over for speeding. During the high speed chase, Vince strikes a pedestrian and kills him. He flees the scene leaving Donna alone in the car. Brought to a women's prison in a tropical country which resembles the film's Philippines-set location, Lee (Pam Grier) and Karen (Margaret Markov) encounter sadistic matron Densmore (Lynn Borden) who voyeuristically observes through a peephole as the inmates take a shower.
The film is a satire of the women in prison film genre and deliberately implements many of the tropes commonly found in such films. Such scenes include nude shower scenes, fight scenes, and a suggested romantic relationship between one of the inmates and an administrator. The overall plot involves a new influx of girls coming to the school. They are immediately confronted with Charlie Chambliss (Williams) who is the de facto leader of the school and has an exceedingly close relationship with the head of the ward, Edna (Ast). Charlie and her circle are given special privileges by Edna and it is suggested that Charlie and Edna enjoy a more intimate relationship. Charlie runs a secret society of girls who are loyal to her and to whom she offers protection. The two main new girls break several of Edna's rules and are punished decisively for their infractions.

Ilsa is Kommandant of a Nazi prison camp, who conducts sadistic scientific experiments designed to demonstrate that women are more capable of enduring pain than men are, and therefore should be allowed to fight in German armed forces (it is late in the war and the Nazi military is in dire need of reinforcements). Ilsa is also portrayed as a buxom woman with a voracious sexual appetite for men. Every night, she chooses another one of her male prisoners and rapes him; however, due to her insatiable hunger, she gets disappointed when her current victim eventually ejaculates, and promptly has him castrated and put to death. Only an American prisoner, who can withhold ejaculating, manages to use her weakness to his favor.
