In the South Indian kingdom of Cholapuram Paalayam in the year 1771, Raja Mokkaiyappar and his queen, Rajamatha Bhavani Ammaiyar are desperate for a child, as all their previous twenty-two children have died at birth. Unknown to them, Bhavani's brother, Sangilimayan, who is also the Rajaguru (Chief Minister), is responsible for the death of their children. Sangilimayan is shocked to see that his sister has given birth to twins. Before informing the king about the birth, he calls the palace astrologer, Chinnavadayaan, who predicts that the elder twin will be incapable of making decisions on his own while the younger one will be smarter. Sangilimayan orders the palace doctor, Kailasakaruppan, to kill the younger one, but Chinnavadayaan tells Sangilimayan that his actions would not be good for the kingdom. Instead, Sangilimayan orders the palace doctor to abandon the child in a nearby river. The elder child is named Pulikecei XXIII. Maragathavalli, Kailasakaruppan's childless wife, rescues the abandoned child from the river, and the couple decides to raise him as their own, naming him Ukraputhan.

The town of Jayshankarapuram, home of Tamil-speaking cowboys, is ruled by a one-eyed tyrant Kelakku Kattai (Nasser) (literally "Eastwood"), who rules over other villages as well, and is always surrounded by his assistants, a cowgirl named Pakki (Lakshmi Rai) and an outlaw named Ulakkai (Sai Kumar). Rasu (Santhanam) and Mokkai ('Ganja' Karuppu) are youngsters sharing a hostel with a diverse group of people including a professor (Madan Bob), a staunch atheist (Rajesh), a poet and other unemployed, struggling men. Rasu, who works in a coffee shop, is in love with Mahishasuramardhini (Madhumita), but she seemingly ignores all his advances. Rasu was even attacked by her brother after hanging around her house for extended periods of time. Mokkai, who is unemployed due to his lack of education, and does whatever job he can find, dreams of marrying his uncle's daughter back in his village. After a particularly bad day where Mokkai is arrested and beaten by female police officers, Rasu's lies of being in a well-paid job are uncovered (to the disgust of Mahishasuramardhini), the two are also kicked out of the hostel by the owner due to the inability to pay rent. On their last night, both vent their frustration at God after heavy drinking. Both are shocked when GOD(Prakashraj) appears in front of them. Mistaking Him for another unemployed fellow, the two head back to their hostel room only to find God waiting for them in it, waving aside this miracle, all three fall asleep. The next morning, God finally manages to convince Rasu and Mokkai of his true identity after appearing in the forms of Vishnu, Jesus and the Buddha.

The film starts with an outsider coming to the village to provide invitations to the people there. There is a police station with one sub-inspector (Bagavathi Perumal) and 3 constables. The village is apparently winning the award from President for being a model village. People are very honest, hard working and there are not problems in the village. The Policemen lead a very peaceful life because of the villagers. They work from 9 to 5 and are closed on sundays. The police station is used for playing board games and watching tv along with the villagers. Constable Shanmugha Pandi (Arulnithi) is a day dreamer and has a liking to a school teacher (Remya Nambeesan).

Prabhu Sivaanu (Siddharth) and his friends discuss the route from Bangalore to Chennai and plan to kidnap Rithika (Ashrita Shetty). Meanwhile her dad appoints encounter specialist Manoj Menon (Kay Kay Menon) who questions Rithika's male friend Deepak (Deepak) about Prabhu. The guy claims that Prabhu is a drug addict and was always trying to woo Rithika, who started to avoid him because of Prabhu. After that the plot continues when Manoj tries to track Prabhu and his friends. In the attempt, they track down a message from another friend Umesh asking Prabhu and friends to come to Bangarpet bus stand only to find Manoj and his team arresting the guy for more information. Manoj goes to a railway station where the train in which Prabhu and Rithika are travelling to Chennai. He finds Rithika and forcefully drags her into his jeep. As the flashback winds again (this time by Rithika), she fell for Prabhu after seeing him on many instances showing his presence of mind. As the flashback ends, Manoj finds Prabhu on the highway where he doesn't stop until he runs over pieces of glass put down by Prabhu. They get into a brawl where Prabhu loses and Manoj continues in the highway but has to stop in a railway signal. Prabhu takes his chance and threatens Manoj keeping his throat under a blade and taking Rithika with him. After a cat and mouse chase, Manoj calls Prabhu saying that his friend needs immediate treatment after an accident. He goes in search of Manoj and involves in a fight with him again. Now, after the stroke of midnight Rithika turns 18 giving her the right for choosing her partner. After that Manoj leaves the Highway as he couldn't do anymore according to law.
