Derek Charles (Idris Elba) works for a finance company and is married to Sharon (Beyoncé Knowles). While Derek is at work, he greets temporary worker Lisa Sheridan (Ali Larter), who—believing Derek was flirting with her—attempts to seduce him throughout the film. Derek repeatedly rejects her, but Lisa continues to pursue him, making sexual advances on him at the Christmas party and flashing him in his car. Derek intends to report Lisa to his firm's human resource management, but learns that she has quit her job. Thinking that Lisa has given up, Derek is annoyed when he receives flirtatious emails from her. Derek and his workmates visit a resort for a conference, where he spots and confronts Lisa, who spikes his drink. Incapacitated, Derek is helpless when Lisa follows him into his hotel room and rapes him. He confronts Lisa again the following day, and hours later discovers her lying naked in his bed after attempting suicide by drug overdose. Derek calls for medical help.

The film tells the story of Lucian, the first werewolf born in human form and the first to be called a Lycan. Viktor, a vampire elder, raises the child, and envisions a race of Lycan slaves that could keep guard of the coven's fortress during the day and be used as laborers by the vampires at night. To this end, Viktor demands from the local human nobles a tribute of silver, with which to keep his slaves under control. As Lucian grows up, he and Viktor's daughter Sonja (Rhona Mitra) fall in love and begin a secretive intimate relationship in their adult years. Sonja is reckless and insubordinate, and one night Lucian escapes and rescues Sonja from his werewolf brethren. Viktor, despite acknowledging that Lucian saved his daughter, cannot forgive the escape and locks Lucian away.

On one side of town, a babysitter and the kids she is babysitting are brutally murdered. The police find that the murderer used his bare hands to tear them to shreds. The shot then changes to teenager Jill Johnson (Camilla Belle) who is going through a teenage crisis, having her boyfriend, Bobby (Brian Geraghty), cheat on her with her best friend Tiffany (Katie Cassidy), and going over 800 minutes on her cell phone, for which her dad, Ben (Clark Gregg), has her babysit for a wealthy family to pay the debt. The unfortunate timing causes Jill to miss out on a school bonfire which her other best friend, Scarlett (Tessa Thompson), is attending. Jill arrives at the elaborate Mandrakis House, which has a greenhouse built in the center. The parents (Derek de Lint and Kate Jennings Grant) show her around and tell her about their live in housemaid Rosa (Rosine Ace Hatem). Mrs Mandrakis then gives Jill their numbers, stating that they won't be back until midnight. As the kids (Arthur Young and Madeline Carroll) are seen to be upstairs asleep, Jill soon begins to receive anonymous phone calls.

Ethan Dulles (who calls himself "Cool Ethan") (Jason Schwartzman), a classmate obsessed with Angela to the extent of collecting loose hairs and making a hair doll and having surveillance photos and a shrine to Angela setup in his dorm room, takes her exam question sheet after Angela leaves and uses it to repeatedly confront and then blackmail the guys into setting up a successful date with Angela for him in exchange for his silence on the matter. The guys set Ethan up in multiple situations in an attempt to convince Angela to like him, while Dave tells Sam during their work researching her that Angela is no more important to him as any other scam they've done. Ethan fails to attract her after frequent confrontations based on his delusional behavior, immaturity and ignorance of social norms.
