Zach Cooper (Dylan Minnette) and his mother Gale (Amy Ryan) move from New York to the fictional town of Madison, Delaware. Their new neighbors are the mysterious "Mr. Shivers" (Jack Black) and his daughter Hannah (Odeya Rush), but when Zach tries to introduce himself, Mr. Shivers warns him to stay away from them. At school, Zach befriends Champ (Ryan Lee), an often bullied, socially awkward student. One night, Zach hears Mr. Shivers and Hannah arguing from his window, followed by Hannah screaming. He calls the police, but Mr. Shivers assures the officers that nothing is wrong and the scream was from a movie. Fearing Hannah is in danger, Zach tricks Mr. Shivers into going to the police station for further questioning.

Will Randall is bitten by a wolf while driving home in Vermont. Afterwards, he is demoted from editor-in-chief of a publishing house when it is taken over by tycoon Raymond Alden, who replaces him with Will's protégé Stewart Swinton. When Will thinks that his wife Charlotte is having an affair with Stewart, he rushes over to his house where Stewart sees Will angry and asks him what the problem is. Will bites Stewart and rushes upstairs to the room to find Charlotte half-naked. His worst fears are confirmed. Will becomes more aggressive, taking on the characteristics of a wolf.

Wilfred Glendon (Henry Hull) is a wealthy and world-renowned English botanist who journeys to Tibet in search of the elusive mariphasa plant. While there, he is attacked and bitten by a creature later revealed to be a werewolf, although he succeeds in acquiring a specimen of the mariphasa. Once back home in London he is approached by a fellow botanist, Dr. Yogami (Warner Oland), who claims to have met him in Tibet while also seeking the mariphasa. Yogami warns Glendon that the bite of a werewolf would cause him to become a werewolf as well, adding that the mariphasa is a temporary antidote for the disease.

Former NYPD Captain Dewey Wilson (Albert Finney) is brought back to the force and assigned to solve a bizarre string of violent murders after high-profile magnate Christopher Van der Veer (Max M. Brown), his wife (Anne Marie Pohtamo), and his bodyguard are slain in Battery Park. Executive Security, Van der Veer's client, prefers to blame the murders on terrorists; but knowing that the victim's bodyguard was a 300-pound Haitian with voodoo ties makes Wilson skeptical. At the crime scene, Wilson meets with Warren (Dick O'Neill), his superior. With pressure to solve the case coming down from both the Police Commissioner and the Mayor, Warren partners Wilson with criminal psychologist Rebecca Neff (Diane Venora). Meanwhile, a homeless man goes exploring an abandoned church in Charlotte Street, South Bronx, which was going to be demolished by Van der Veer along with the rest of the ruined buildings in the area, to be able to build apartment complexes. He is suddenly attacked and torn to pieces by an unseen monstrous being. Wilson and Neff arrive to investigate his murder. While investigating the abandoned church, sounds of crying lure Neff up the bell tower. Wilson follows her but doesn't hear the crying; however, once Neff is separated from him, he hears a wolf howl. He goes up after Neff and drags her forcibly to safety. Later, during the night, a bridge worker is apparently murdered by the same creature.

Dracula seduces Hussmann's granddaughter-in-law Rita (Anne Gwynne) and kills Hussmann himself, but in a subsequent chase, Niemann disposes of Dracula's coffin, causing the vampire to perish in sunlight.

While at a party, Scott keeps undergoing changes and eventually he returns home, locks himself in the bathroom, and undergoes a complete change and becomes a werewolf, while his father demands that he open the door. He tries to refuse, only to finally give in and find his father has also transformed. Harold never told his son about the condition because "sometimes it skips a generation" and he was hoping it would not happen to Scott. Needing someone else to share his secret, Scott confides his best friend, Stiles Stilinski (Jerry Levine), who accepts Scott's werewolf status. However, Scott inadvertently reveals his transformation to the public at one of his basketball games. After shortly stunning the crowd with The Wolf, Scott goes on to amaze them with his newfound basketball skills.

In a small suburban town, a group of university students--Mark Loftmore (Zach Galligan), China Webster (Michelle Johnson), Sarah Brightman (Deborah Foreman), Gemma (Clare Carey), James (Eric Brown) and Tony (Dana Ashbrook)--visit a mysterious wax museum, where they encounter several morbid displays, all of which contain stock characters from the horror genre. Tony and China unintentionally enter two separate pocket worlds, as depicted by the waxwork displays, by crossing the exhibition barrier rope. Tony is at a cabin where a werewolf (John Rhys-Davies) attacks him. A hunter and his son arrive and try to kill the werewolf. The son fails and is torn in two, while the hunter shoots the werewolf, and then shoots Tony as he begins to transform into a werewolf. China is sent to a Gothic castle, where vampires attack her and Count Dracula (Miles O'Keeffe) turns her into a vampire. Two of the other students, Mark and Sarah, leave the waxwork unscathed. Later, Jonathan (Micah Grant), "a college jock", arrives at the wax museum, looking for China, but The Phantom of the Opera display gets his attention as David Lincoln (David Warner) walks him into the display. Mark goes to a pair of investigating police detectives, Inspector Roberts (Charles McCaughan), and meets Lincoln as he lets Roberts investigate the waxworks. As Mark and Roberts leave the Museum, Mark recognizes Lincoln.
