In New York Harbor, Carl Denham, famous for making wildlife films in remote and exotic locations, charters Captain Englehorn's ship Venture for his new project, but he is unable to secure an actress for a female role he reluctantly added. Denham searches the streets of New York for a suitable woman. He meets penniless Ann Darrow and convinces her to join him for the adventure of a lifetime. The Venture quickly gets underway. The surly first mate, Jack Driscoll, gradually falls in love with Ann. After weeks of secrecy, Denham finally tells Englehorn and Driscoll that their destination is Skull Island, an uncharted island shown on a map in Denham's possession. Denham speaks of something monstrous there, a legendary entity known only as "Kong".

The story picks up about a month after the dramatic finale of the previous film and follows the further adventures of filmmaker Carl Denham, now implicated in numerous lawsuits following the destruction wrought by Kong. Denham leaves New York with the captain of the Venture, Captain Englehorn, who is certain it is just a matter of time before he is similarly served. Their efforts to make money shipping cargo around the Orient are less than successful. In the Dutch port of Dakang, Denham is amused to see there's a "show" being presented, so he and Englehorn attend. It turns out to be a series of performing monkeys, capped by a song ("Runaway Blues") sung by a young woman named Hilda.

From a lost expedition to a plateau in Venezuela, Paula White brings the journal of her father explorer Maple White to the eccentric Professor Challenger in London. The journal features sketches of dinosaurs which is enough proof for Challenger to publicly announce that dinosaurs still walk the earth. Met with ridicule at an academic meeting at the Zoological Hall, Challenger reluctantly accepts a newspaper's offer to finance a mission to rescue Maple White. Professor Challenger, Paula White, sportsman Sir John Roxton, news reporter Edward Malone (who is a friend of Roxton and wishes to go on the expedition to impress his fiancée), a sceptical professor Summerlee, an Indian servant Zambo, and Challenger's butler Austin leave for the plateau.

« Qu’en était-il du triste sort infligé aux scénaristes à l’époque des cavernes ? Jugez par vous-mêmes ». Un personnage chauve et à lunettes, assis dans son bureau, producteur de la préhistoire — reconnaissable à son short en peau de bête — reçoit une proposition de script sous forme d’un rouleau de parchemin qu’il commence à réduire à grands coups de ciseaux. Il auditionne d’abord une jeune femme qui se livre à un numéro de charme se terminant par un grand écart, puis il reçoit un clown coiffé d’un chapeau melon (qui pourrait être Charlot), avant d’engager un athlète qui lui montre sa force en malmenant une chèvre que l’on a remarquée auparavant broutant le short du producteur et les pages refusées. « Et maintenant, ils tournent ce qui reste de l’histoire ». On assiste à une querelle entre l’athlète et celui qui doit être le réalisateur, qui se termine par un K.O. général. Puis la vedette, chevauchant la chèvre comme un pur-sang, galope dans la nature, suivi par un dinosaure qui sert de grue pour effectuer un mouvement de caméra, l’appareil de prise de vues étant très drôlement actionné par le caméraman qui fait tourner un pédalier de vélo et non pas une manivelle. L’homme au petit chapeau s’exerce ensuite au lancer de couteaux sur l’athlète, ligoté par un singe, puis fait la cour à la belle. Le couple reçoit enfin la bénédiction d’un chaman perché en haut d’un cocotier. Le film doit encore subir les outrages de la censure, représentée ici par trois hommes à chapeau haut-de-forme qui menacent le producteur à qui il ne reste plus qu’à mutiler la pellicule. « Et c’est alors que s’avance le fier scénariste pour voir ce qu’on a fait de son chef d’œuvre ». La projection commence avec le titre à double sens : « Qui est le bouc ? bâclé d’après l’histoire d’Aloysius Flintpebble ("pierre à silex")». Film qui a obtenu le « visa N° 7-11 » ! Il ne reste de l’histoire filmée que des débris sans queue ni tête mais qui se termine par un mariage. Le scénariste s’arrache les cheveux et met au tapis le producteur. « C’est un bon scénario qui reconnaît son auteur… Après, ça devient un film » (It’s a wise scenario that knows its own author… after it gets in the movies).

It starts with a caveman going to give some flowers to a cavegirl. He fails when he hits a tree. However, he keeps going. However, the "Self Appointed Hero" of the story steals the girl's heart. Meanwhile, an evil Gorilla-like creature called "Wild Willie" the Missing Link is watching them. When the Missing Link goes to the lake to get some water, where the dinosaur is, the dinosaur kills the Missing Link and goes away. Then the "Hero" finds the Missing Link and takes the credit for killing Wild Willie.

When her master McCay calls her, the frisky, childlike Gertie appears from a cave. Her whip-wielding master has her do tricks such as raising her foot or bowing on command. When she feels she has been pushed too far, she nips back at her master. She cries when he scolds her, and he placates her with a pumpkin. Throughout the act, prehistoric denizens such as a flying lizard continually distract Gertie. She tosses a mammoth in the lake; when it teases her by spraying her with water, she hurls a boulder at it as it swims away. After she quenches her thirst by draining the lake, McCay has her carry him offstage while he bows to the audience.
