Nate (Mickey Rourke), a small-time jazz musician and recovering heroin addict, is clearly a hard-luck case. After a performance one night, he is mugged and bound. He awakens to find himself in a vehicle and being driven out into the desert, but he is helpless to do anything about it. His assailant then walks Nate away from the car, and is about to kill him, but instead the assailant is shot dead by a band of Native Americans, and Nate is left unharmed. Nate wanders around in the desert and climbs a mountain, where he looks down upon a carnival and sideshow. He walks through the carnival, asking the owner to use a telephone, and he wanders among the fire eaters, sword swallowers, and other performers. Nate finds himself drawn to the exotic beauty of Lily (Megan Fox), a freak performer who has wings on her back. Lily is cold and dismissive, but it dawns on her that this gentle giant may well be her ticket to a better life. The sideshow owner (Rhys Ifans) attempts to kill Nate, but Lily rescues him by stealing a truck, and together they flee to the city.

Sixteen-year-old Minnesota high-schooler Juno MacGuff (Ellen Page) discovers she is pregnant with a child fathered by her friend and longtime admirer, Paulie Bleeker (Michael Cera). She initially considers an abortion. Going to a local clinic run by a women's group, she encounters outside a schoolmate who is holding a rather pathetic one-person pro-life vigil. Once inside, however, Juno is alienated by the clinic staff's authoritarian and bureaucratic attitudes. She is particularly offended by their calling her "sexually active", a term which she feels demeans the highly emotional event by which she became pregnant. All of this decides her against abortion, and she decides to give the baby up for adoption. With the help of her friend Leah (Olivia Thirlby), Juno searches the ads in the Pennysaver and finds a couple she feels will provide a suitable home. She tells her parents, Mac (J.K. Simmons) and stepmother, Bren (Allison Janney), who offer their support. With Mac, Juno meets the couple, Mark and Vanessa Loring (Jason Bateman and Jennifer Garner), in their expensive home and agrees to a closed adoption.

Nick Naylor is a handsome, smooth-talking tobacco lobbyist and the vice-president of a tobacco lobby called the "Academy of Tobacco Studies", which for 15 years has been "researching" the link between tobacco smoking and lung cancer. They claim that their research—funded primarily by tobacco companies—has found no definitive evidence of any linkage. Naylor's job consists mainly of reporting the questionable research of the "Academy" to the public and defending Big Tobacco on television programs by questioning opposing health claims and advocating personal choice. Naylor and his friends, firearm lobbyist Bobby Jay Bliss and alcohol lobbyist Polly Bailey, meet every week and jokingly call themselves the "Merchants of Death" or "The MOD Squad".

A young woman, Alice (Selma Blair), goes to the gynecologist with a mysterious ache inside her. When one doctor can't figure out just what the problem is, she sees another, and still more, until finally one specialist finds that she has an enormous, empty tundra inside of her, the opening of which is her vagina. She is forced into a media parade, brought onto talk shows so that the specialist can talk about his discovery. Alice is treated coldly, like her emptiness is the only thing important about her, and still she aches. A challenge is put forth to see who would be willing to enter the vast tundra and see where it might lead; several men enter and travel for a long time, but find nothing but ice. One young man enters and does not come back out. At the end, Alice goes to a beach and stands by the water. In a rush, the ice from the emptiness inside her flows out, freezing the ocean, and the young man appears beside her.
