Set in 1910, the film's main character is Ben Harvey (patterned after Ben Hecht): serious about seeing the world, he leaves his home for Chicago, where he meets a woman named Lil, who in reality is the Madam of the bordello Ben mistakes for a boarding house. He also is friends with Adeline, one of the prostitutes. While he tries to find work, Ben encounters other people, including a man named Sullivan, who is involved in shady doings in city government. Suspecting corruption, Harvey and a hard drinking reporter decide to investigate.

While the warden (real-life warden Frank A. Millionaire businessman-sportsman Thomas Crown (Steve McQueen) pulls off a perfect crime by orchestrating four men to rob $2,660,527.62 from a Boston bank, along with a fifth man who drives the getaway Ford station wagon with the money and dumps it in a cemetery trash can. None of the men ever meets Crown face-to-face, nor do they know or meet each other before the robbery. Crown retrieves the money from the trash can personally after secretly following the driver of the station wagon, then personally deposits the money into an anonymous Swiss bank account in Geneva, making several trips, never depositing the money all at once so as to not draw undue attention to his actions.

Mr. Colbert, a wealthy man from Chicago who was planning to build a factory in Sparta, Mississippi, is found murdered. White police Chief Bill Gillespie (Rod Steiger) comes under pressure to quickly find his killer. African-American northerner Virgil Tibbs (Sidney Poitier), passing through town, is picked up at the train station between trains with a substantial amount of cash in his wallet. Gillespie, prejudiced against blacks, jumps to the conclusion that he has his culprit but is embarrassed to learn that Tibbs is an experienced Philadelphia homicide detective who is simply passing through town after visiting his mother. After the racist treatment that he receives, Tibbs wants nothing more than to leave as quickly as possible, but his own chief, after questioning whether Tibbs himself is prejudiced, has him stay and help. Leslie Colbert (Lee Grant), the victim's widow, already frustrated by the ineptitude of the local police, is impressed by Tibbs's expertise when he clears another wrongly accused suspect whom Gillespie has arrested on circumstantial evidence. She threatens to stop construction on the much needed factory unless Tibbs leads the investigation. Unwilling to accept help, but under orders from the town's mayor, Gillespie talks a reluctant Tibbs into working on the case.

Joanna Drayton's unannounced early return from a Hawaii holiday causes a stir when she brings to her childhood upper-class home her new fiancé, John: a widowed, black physician. Joanna's parents - newspaper publisher Matt Drayton (Spencer Tracy) and his wife, art gallery owner Christina Drayton (Katharine Hepburn) – are liberals who have instilled in her the idea of racial equality. But to her surprise, Joanna's parents are deeply upset that she is planning to marry a black man. The Draytons' black maid, Tille (Isabel Sanford), is just as horrified, suspecting that John is trying to "get above himself" by marrying a white woman. Rancher Taw Jackson (John Wayne) returns to his hometown to settle a score. Three years earlier, he was framed by corrupt businessman Frank Pierce (Bruce Cabot) and wrongfully imprisoned. Pierce did this to confiscate Jackson's land, where he had discovered gold. After his prison sentence was cut short for good behavior, Jackson returns to steal a shipment of gold from Pierce. He hires Lomax (Kirk Douglas) to assist him, even though he had worked as a hired gun for Pierce and was instrumental in sending him to prison. Jackson needs Lomax not only because of his marksmanship, but also because he is a safe-cracker. The gold shipment is being transported in a "war wagon," a heavily armored stagecoach armed with a deadly Gatling gun in a top-mounted steerable turret. Jackson and Lomax assemble a gang and plan to rob the war wagon at the weakest point in the route it travels.

The story follows Prince Keoki Kanakoa, the Reverend Abner Hale and his wife Jerusha, who join the prince on mission to the Islands of Hawaii for the Calvinist church with the promise to make Kanakoa a minister. Upon their arrival to the islands, the ship stops in Lahaina, Maui before going on to Honolulu, Oahu where the main church has been established. There they are greeted by the aliʻi nui, Malama Kanakoa, Keoki's mother, who stops the Hales from traveling further when she demands Jerusha remain in Lahaina to teach her to write. Reverend Hale attempts to teach her of the Christian God first but she refuses until she, herself can write. Only after that will she listen to Abner's Christian teachings. As Hale learns, there is a serious difference between a destiny calling to him and the call to recognize the needs of others.
