The film follows three aspiring screenwriters as they struggle to turn their scripts into movies. David is a hip talent agent's assistant with three scripts circulating around town. He's plugged into "young Hollywood" - and when he's not working or writing, he's usually hanging out at the beach. Joe is a middle-aged family man who has split time over the last three years between caring for his autistic daughter and writing what he believes could be the great American screenplay. And Deborah is trying to become one of the few African-American women to ever write and direct a feature film, though she's struggling just to pay her bills while she searches for money to produce her script. Between these stories, the film intercuts critical insight from such Hollywood screenwriters as James L. Brooks, Nora Ephron, Carrie Fisher, Gary Ross, Steven E. de Souza, Ed Solomon, Paul Guay, Scott Alexander and Larry Karaszewski.

Willard (Norm Macdonald) is an overworked, underpaid chauffeur who works for a mean-spirited pie heiress named Mrs. Crock (Elaine Stritch), just as his father did before him. All Willard wants for Christmas is a new uniform, as the one he currently wears is the one his father was buried in, but Crock gives him a cheap pair of cuff links and a pie instead, while lavishing expensive gifts on her business partner Chip Oswald (Sherman Hemsley) and her prized dog Muffin. Finally fed up with being mistreated, Willard and his best friend, local chicken restaurant owner Rusty (Dave Chappelle), concoct a scheme to kidnap the dog and hold it for a $1,000,000 ransom. Muffin attacks him, leaving a great deal of destruction and Willard's blood at the scene, and the plan fails when the dog later escapes.

In the summer of 2006, Percy recounts the story of Thalia and her sacrifice. A young Annabeth, Luke, Grover and Thalia are running to Camp Half-Blood while being pursued by monsters. Thalia sacrifices herself to get the others into the camp, and her father Zeus transforms her into a pine tree, creating a magical border around the camp. Soon a Cyclops passes through the border and is revealed to be Tyson, Percy's half-brother. The camp is later attacked by a Colchis Bull who breaks through the border and ravages Camp Half-Blood. Percy defeats it with the help of Tyson, Annabeth, and Clarisse. Mike Enslin (John Cusack) is a cynical, skeptical author who, after the death of his daughter Katie (Jasmine Jessica Anthony), writes books appraising supernatural events in which he has no belief. After his latest book, he receives an anonymous postcard depicting The Dolphin, a hotel on Lexington Avenue in New York City bearing the message, "Don't enter 1408." Viewing this as a challenge, Mike forces the hotel to allow him to book the room, referencing a law that any hotel room in New York can be requested as long as it meets safety standards. The hotel manager, Gerald Olin (Samuel L. Jackson) tries to dissuade Mike from checking into the room, explaining that 56 people have died in the room over the past 95 years, and that no one has lasted more than an hour inside it. Mike, who does not believe in the paranormal, insists on staying in the room, and asks Olin if he thinks it is haunted; Olin replies that it is "evil" rather than haunted.

In 1952, 10-year-old Larry Flynt is selling moonshine in Kentucky. Twenty years later, Flynt and his younger brother, Jimmy, run the Hustler Go-Go club in Cincinnati. In 1952, Ed Wood (Johnny Depp) is struggling to join the film industry. Upon hearing of an announcement in Variety magazine that producer George Weiss (Mike Starr) is trying to purchase Christine Jorgensen's life story, Ed is inspired to meet Weiss in person. Weiss explains that Variety 's announcement was a news leak, and it is impossible to purchase Jorgensen's rights. The producer decides to "fictionalize" the film, titled I Changed My Sex!. Ed tries to convince Weiss that he is perfect to direct I Changed My Sex! because he is a transvestite, but is unsuccessful since Weiss wants a director with experience. One day, Ed meets his longtime idol Bela Lugosi (Martin Landau), after spotting him trying out a casket. Ed drives Bela home and the two become friends. Later, Wood persuades Weiss to let him direct I Changed My Sex! by convincing him that having a star in the film would sell tickets, and they could sign Bela for a low price.
