Synopsis
En Amazonie, des anthropologues découvrent les restes d'un homme âgé de plus de 300 ans lors de sa mort ! Un expédition est organisée pour trouver le mystère de cette incroyable longévité. Mais au coeur de la forêt impénétrable, une tribu sauvage et un serpent géant à cinq têtes montent la garde... Suite à la découverte d'un tombeau renfermant les restes d'un chef de tribu d'Amazonie, les chercheurs d'une industrie pharmaceutique déterminent l'âge du défunt à environ 300 ans. Pour le monde de la science, ce détail pourrait changer le cours de l'humanité. C'est pourquoi une seconde expédition est envoyée en Amérique du Sud pour percer le secret de cette jeunesse éternelle. The film takes place in Venice, where David Franks (Stephen Baldwin) and his girlfriend Laura (Vanessa Johansson) are meeting with Venetian police to learn more about the mysterious disappearance of David's father. During the boatride to where they think David's father may have last been seen, David sees the dorsal fin of a large shark. In a double-take he looks back and the fin is gone. David and another man (Rossi) enter the canals wearing wetsuits and scuba gear. A large shark preys on the two and in a sudden attack the Italian man is eaten. David is bitten in the shoulder but manages to escape to a cave.

A team of researchers funded by a Manhattan pharmaceutical firm – Jack Byron, Gordon Mitchell, Sam Rogers, Gail Stern, Cole Burris, and Ben Douglas – leave for a jungle in Borneo to search for a "blood orchid", a flower they believe can be used as a type of fountain of youth. Though their guide Bill Johnson has misgivings about which path to take, Jack bribes him to take an unsafe path. The team goes over a waterfall and has to wade through the river. A giant anaconda emerges from the water and swallows Ben whole, but the rest of the team escapes the river. Bill assures them that it was the largest snake he has ever seen and that it should take weeks for it to grow hungry again; however, most of the team demands that the expedition be called off. They travel to Bill's friend, John Livingston, who lives on the river to see if Johnson can borrow his boat; however, they find Livingston dead and his boat crashed.

Mongolia, 1934. The researcher Maple White (Jack Langedijk), together with his assistant Azbek (Russell Yuen), discovers an unknown world populated by dinosaurs, situated on a plateau in Mongolia. After having found a dinosaur's egg in a cave, White celebrates the discovery, with Azbek, uncorking a bottle of champagne and listening to classical music. Azbek is attacked and killed by Dimorphodons, which also attack White who falls from the plateau holding onto a rope; he survives but is seriously wounded, White is found again and taken care of by Myar and Djena (Gregorian Minot Payeur), respectively Azbek's brother and sister; White asks the two to bring him his friend George Challenger (Patrick Bergin) to whom White gives an account of his adventures, proposing to Challenger that he should try to fully explore the plateau in his place.

A baby anaconda, which was captured at the end of the previous film, is being used for experiments by a man named Peter Reysner from the previous film. Peter creates a hybrid of the blood orchids from Borneo that enabled the anaconda to grow so large and live so long, and creates a serum for cell regeneration. After the test seemingly work for the baby anaconda to regenerate, Peter burns it. When he disappears, the long anaconda escapes from the cage and kills Peter in a mine filled with blood orchids. J.D. Murdoch, a billionaire suffering from bone cancer, sends his assassin Eugene and his team of henchmen mercenaries to find Peter and the serum of Peter created it that can cure him. He also tells them to be careful of Dr. Amanda Hayes, and to kill her if needed. As Dr. Amanda Hayes, the sole survivor of the previous snake attack, and two officers also go in search of Peter, determined to destroy the serum and kill the snake.

The crew of a plane gets nervous when the cargo it is carrying starts making noises and shifting. When one of the crew opens the crate, the pilot hears screams from the back. In Romania, a female anaconda and a male anaconda captured, are being held in a genetic research facility, Wexel Hall for experimentation. The research project is led by Dr. Amanda Hayes (Crystal Allen) and funded by J.D. Murdoch (John Rhys-Davies), a well-known industrialist. While visiting the facility, Murdoch agitates the male anaconda with a large flashlight, and has it gassed to calm it down. However, as Murdoch, his assistant Pinkus (Ryan McCluskey) and a scientist leave, the anaconda breaks through his enclosure's wall. As it escapes, it kills many of the people working in the lab, though Cupboard escapes the destruction. In looking through the facility, Amanda and another researcher realize that the second (Queen) anaconda has also escaped. Murdoch calls in a team of animal hunters, led by Hammett (David Hasselhoff), to capture both snakes. Amanda and Pinkus go with them. At a small farm in the middle of the woods, the owner is eaten alive by one of the snakes.

A group of teenagers journey to a remote jungle in search of a friend who's been reported as lost after his trip to the jungle. They use a hot air balloon to search for him. While searching, they are caught in a brutal wind storm which blows their vessel into the middle of the jungle.
