The film follows three groups of people, all of whom have found a video camera on their doorstep and begin filming under the impression that this is the key to winning money from a mysterious competition. Tom (Todd Stashwick) is an average guy filming the life of his family with his wife Emmy. While Beth (Alexandra Lydon) is a bored and isolated college girl who sees the camera as something to fill her free time. Meanwhile, Leonard (Barak Hardley) is a mother's boy who believes his clown makeup will steal the scene. Each group has been given a label - "The Family" (Tom & Emmy), "The Woman" (Beth), and "The Clown" (Leonard), but they are largely unaware of what is truly going on and are shocked when they receive instructions telling them to keep filming.

Nothing much happens in the lives of 20-something pals Dexter and Royce except for getting high and hanging out with Royce’s girlfriend, Matilda ('Mattie'). This all changes one evening in Northern Ontario town Weedsville when Mattie overdoses on a stash of Dexter and Royce’s drugs — drugs fronted by local drug kingpin and tough-guy Omar to sell in order to cover their previous drug debt. Ann (Sarah Polley) is a hard-working 23-year-old mother with two small daughters, an unemployed husband (Scott Speedman), a mother (Deborah Harry) who sees her life as a failure, and a jailed father whom she has not seen in ten years. Her life changes dramatically when, during a medical checkup following a collapse, she is diagnosed with metastatic ovarian cancer and told she has only two months to live. Deciding not to tell anyone of her condition, using the cover of anemia, Ann makes a list of things to do before she dies. She decides to change her hair, record birthday messages for the girls for every year until they're 18, and tries to set up her husband with another woman. Feeling a longing to experience a life that was never available to her, she seeks out a man to experience how it feels to be in a sexual relationship with someone other than her husband. Her experiment ends up taking an emotional toll when she meets with a man named Lee, who ends up madly in love with her and is left heartbroken when Ann breaks up. He meets with her one last time and says that he will do anything to make her happy, taking care of her daughters and even finding her husband a job. She ends their relationship and never tells him that she is dying. At the end of the film Ann records a message to her husband telling him that she loves him and another one to Lee telling him the same. She then leaves all tapes she has recorded with her doctor asking him to deliver them after her death.

Laurel and Audrey are identical twins. Although they look the same on the outside, their personalities are very different. When they get in a severe car crash together, Laurel is taken to the hospital and is informed that her sister has died. Suffering from post-traumatic amnesia, Laurel doesn't remember being who she is at first. The morning of her sister's funeral, she remembers who she is but decides to keep everyone thinking that it was she who had died and not Audrey. She then flies back to the city where Audrey used to live. There, she meets her sister's tenant Basel (Jake Johnson) who is confused about her sudden change of personality, because Audrey was never nice to him before. Living her life posing as her sister, she learns that Audrey had a boyfriend who is married, but broke up with him before the accident. Laurel continues to spend more time with Basel and Audrey's best friend Claudia, who is suspicious about Audrey's change of behavior. Laurel begins a relationship with Basel, and they fall in love. When she finally feels that she is adapting to her new life, she accidentally tells her boss that Charles is her ex-boyfriend, not knowing that Charles is actually her boss's husband, and gets fired immediately. Shortly after this, Basel proposes to Laurel saying that he loved her since he first met her. Laurel, unable to continue deceiving everyone, decides to tell Basel the truth, but he is devastated and breaks up with her.
