The story starts out with the regular lives of three friends—Dave (Barry Watson), Adam (Michael Rosenbaum), and Doofer (Harland Williams)—who are head of the Social Committee in a frat house called KOK (Kappa Omicron Kappa). Jennifer Verti, has spent all of her savings buying her first house and all of her belongings have recently been stolen while trying to move into her new house. Vince is a ladies' man and a gambler, and owes money to a debt collector known as "The Monkey" (real name is Eugene). Berke Landers (Ben Foster) and his girlfriend Allison (Melissa Sagemiller) were the quintessential high-school couple who grew up together and eventually fell in love, but she breaks up with him immediately after the film begins. This leads to an opening musical number of "Love Will Keep Us Together" by Vitamin C, imagined by Berke. He seeks advice from his embarrassing parents Frank (Ed Begley Jr.) and Beverly Landers (Swoosie Kurtz), who are hosts of a relationship advice show called Love Matters, but they don't help with the situation and constantly focus on his sex life and sexuality throughout the film. Allison then starts a relationship with Striker (Shane West), a 'foreign' student who was once the lead singer of a boy band called "The Swingtown Lads". When Allison and Striker audition for the school's upcoming musical, Berke desperately tries to win Allison back by also auditioning for the play, despite having no theatrical talent and having a busy schedule as a member of the basketball team. Meanwhile, Berke's friends Felix (Colin Hanks) and Dennis (Sisqó) try to find a new girlfriend for him.

A girl named Riley is born in Minnesota, and within her mind, five personifications of her core emotions—Joy, Sadness, Disgust, Fear, and Anger—come to life. The emotions live in Headquarters, Riley's conscious mind, where they influence Riley's actions and memories via a control console. Her new memories are housed in colored orbs, which are sent into storage at the end of every waking period. The most important or "core" memories are housed in a hub in Headquarters and power five "islands", each of which reflects a different aspect of Riley's personality. Joy acts as the dominant emotion to keep Riley in a happy state, but she and the others do not understand Sadness's purpose, other than making Riley cry.
