The story starts out with the regular lives of three friends—Dave (Barry Watson), Adam (Michael Rosenbaum), and Doofer (Harland Williams)—who are head of the Social Committee in a frat house called KOK (Kappa Omicron Kappa). Jennifer Verti, has spent all of her savings buying her first house and all of her belongings have recently been stolen while trying to move into her new house. Vince is a ladies' man and a gambler, and owes money to a debt collector known as "The Monkey" (real name is Eugene). They meet at a wedding, initially Jennifer dislikes Vince and begins to predict what kind of person he is, and as a result, he dislikes her. However they talk about weddings and are amazed by the cash and gifts a couple get, and consider how easy it would be get that kind of income. Berke Landers (Ben Foster) and his girlfriend Allison (Melissa Sagemiller) were the quintessential high-school couple who grew up together and eventually fell in love, but she breaks up with him immediately after the film begins. This leads to an opening musical number of "Love Will Keep Us Together" by Vitamin C, imagined by Berke. He seeks advice from his embarrassing parents Frank (Ed Begley Jr.) and Beverly Landers (Swoosie Kurtz), who are hosts of a relationship advice show called Love Matters, but they don't help with the situation and constantly focus on his sex life and sexuality throughout the film. Allison then starts a relationship with Striker (Shane West), a 'foreign' student who was once the lead singer of a boy band called "The Swingtown Lads". When Allison and Striker audition for the school's upcoming musical, Berke desperately tries to win Allison back by also auditioning for the play, despite having no theatrical talent and having a busy schedule as a member of the basketball team. Meanwhile, Berke's friends Felix (Colin Hanks) and Dennis (Sisqó) try to find a new girlfriend for him.

Amir is a professional heartbreaker (people hire him to break up with their boyfriend/girlfriend when they don't want to do it themselves). At Amir's 30th birthday party, his childhood friend Charlie announces that he is getting married, but he and his fiance decide they don't want Amir to be the best man because of his bad "relationship karma". This depresses him, as do the relationship advice books he receives as gifts. Amir goes to visit his family for Thanksgiving and meets Zara, his attractive British cousin, who he hasn't seen in twenty years. Amir's mother persuades him to take Zara back to her home in Southern California, since she is in the United States traveling.
