One night, a thief fleeing through Georgetown in Washington, D.C., is shot by a man carrying a briefcase. A pizza delivery man who witnesses the incident is also shot by the killer and is left in a coma. The following morning, a young woman is killed by a Washington Metro train in what seems to be suicide. Congressman Stephen Collins (Ben Affleck) is distraught to hear that the woman was Sonia Baker (Maria Thayer), a lead researcher on his staff. Collins, who has military experience, is leading an investigation into PointCorp, a private defense contractor with controversial operations involving mercenaries. Collins tells his former college roommate and old friend Cal McAffrey (Russell Crowe), an investigative reporter, that he had been having an affair with Sonia and that she had sent him a cheerful video message on the morning of her death, which he says is inconsistent and unusual behavior for someone about to commit suicide.

In August 1884, Dan Evans (Bale) is an impoverished rancher and Civil War veteran. He owes money to Glen Hollander (Loftin) and when he fails to pay, two of Hollander's men set his barn on fire. Private investigator Patrick Kenzie and his partner/girlfriend Angie Gennaro witness a televised plea by a woman named Helene McCready for the return of her missing daughter Amanda, who was abducted with her favorite doll "Mirabelle". Patrick and Angie are then hired by the child's aunt Beatrice to find Amanda and discover that Helene and her boyfriend "Skinny Ray" had recently stolen money from Cheese, a local Haitian drug lord. After Ray is murdered, Patrick and Angie join the police detectives investigating the case, Remy Bressant and Nick Poole, to arrange a trade of the money for Amanda. Captain Jack Doyle reads Patrick a telephone transcript of the drug lord setting up an exchange for Amanda. The exchange at a nearby quarry in Quincy is botched and Amanda is believed to have drowned, as her doll is found in the quarry and returned to Helene. Doyle, whose own daughter was killed years before, takes responsibility for the death and goes into early retirement.

In 1974, Ronald DeFeo, Jr. murdered his family at their house in Amityville, New York. He claimed that he was persuaded to kill them by voices he heard in the house. In 1974, Ron Burgundy (Will Ferrell) is the famous anchorman for a local San Diego television station, fictional KVWN channel 4. The NSA-funded QT (Quantum Tech) Corporation has slated a project to develop Hypertime, a technology which allows the user's molecules to speed up to the point where the world appears to be standing still. After realizing that such technology, contained within a wristwatch frame, could also be used against the USA, the NSA orders the project stopped. However, the research is farther along than the NSA expected and QT's leader Henry Gates plans on using the technology to usurp the leader of the NSA and dominate the world. He uses the prototype to stretch the weekend in order to give the brilliant lead scientist Earl Dopler time to fix the remaining glitch in the technology after his henchmen Richard and Jay prevent Earl's incognito departure at the airport. However, the disadvantage of Earl being in Hypertime for too long was him aging rapidly in real time, as his molecular age continued at the same rate despite time slowing down.

A couple checks into a suite in Las Vegas. In flashbacks we see that he's a computer whiz on the verge of becoming a dot.com millionaire (Peter Sarsgaard), she's a lap dancer at a club (Molly Parker). He's depressed, withdrawing from work, missing meetings with investors. He wants a connection, so he offers her $10,000 to spend three nights with him in Vegas, and she accepts with conditions: four hours per night of erotic play, and no penetration. During the days in Vegas, they get to know each other, have fun, meet a friend of hers, casino dealer Jerri (Carla Gugino); at night, at least after the first night, things seem to get complicated. When the three days are over, the stripper makes it clear that she was only there for the money and that the man she spent the time with was just a client. Upset that the feelings he had weren't mutual he then rapes her, which she makes no attempt to stop. She then masturbates for him to achieve orgasm. The next day he returns home heartbroken. The movie ends with his return to the strip club to see the woman he fell in love with again. She greets him fondly and says that she is glad to see him again. Because the movie is shown in a non-linear format, it is left to the viewer's interpretation of when this event occurred.
