In an unnamed Latin American dictatorship, a group of rebel freedom fighters fight to bring about a less oppressive society. The rebels, led by the enigmatic Enrique Carrasco (Collins) who has returned to his native land to fight, attack a hydro-electric dam. Along with Carrasco's native fighters are Smithy (Steiner), a British mercenary, and Maria, a native ex medical-student turned freedom fighter. After killing the security guards, Carrasco's men place explosives at a crucial point in the dam wall, when they blow the dam the resulting tidal surge destroys a nearby bridge that is carrying Government convoy.

The film opens with a flashback to the Vietnam War, where Norman Hopper (Saxon) is bitten by a U.S. POW Charlie Bukowski (John Morghen), who is infected with a virus which leaves people with a craving for human flesh. In Atlanta, Georgia, some years later, Hopper wakes up from a nightmare about this incident, and then receives a phone call from Bukowski, who invites his old comrade out for a drink. The call came in at an inopportune moment, as a young neighbour girl, Mary (Cindy Hamilton), was trying to seduce him, so he turns down the invitation. Hopper falls for her charms and as they are about to engage in oral sex, he bites her. After hearing from his concerned wife that a Vietnam vet barricaded himself in a mall, he gets into his car. Just as he is about to leave, Mary reveals that she enjoyed the bite. After arriving on the scene, he convinces Bukowski to surrender to the police. While being hauled away to the hospital, Bukowski bites a police constable.

Martial arts expert Ho Chiang journeys to America's Wild West in order to retrieve his late uncle Wang's missing fortune. Ho must free his uncle's murderer, Dakota (frequent Spaghetti Western star Lee Van Cleef), the only man who knows Wang's final resting place. Retired safe-cracker Chris Gretchko is coaxed out of comfortable retirement by Jeff Olafson, the impulsive son of a former accomplice, who begs him to help steal an undocumented shipment of diamonds to satiate Van Stratten, a New York City crime boss whose mistress, Jessica, has been seeing Jeff on the side. In 1560, several score of Spanish conquistadors, and a hundred Indian slaves, march down from the newly conquered Inca Empire in the Andes mountains into the jungles to the east, in search of the fabled country of El Dorado. Under the command of Gonzalo Pizarro (Alejandro Repullés), the men, clad in half armor, pull cannons down narrow mountainous paths and through dense, muddy jungle. On New Year's Eve, reaching the end of his supplies and unable to go on without more information, Pizarro orders a group of forty men to scout ahead by raft down river. If they do not return to the main party within one week with news of what lies beyond, they will be considered lost. Pizarro chooses Don Pedro de Ursúa (Ruy Guerra) as the commander of the expedition, Don Lope de Aguirre (Klaus Kinski) as his second-in-command, fat nobleman Don Fernando de Guzmán (Peter Berling) representing The Royal House of Spain and Brother Gaspar de Carvajal (Del Negro) to bring the word of God. Also accompanying the expedition, against Pizarro's better judgment, are Ursúa's mistress, Doña Inés (Helena Rojo) and Aguirre's teenage daughter, Florés (Cecilia Rivera, in her only film role).
