The film opens with homemade videos of fraternal twins Jack and Jill Sadelstein growing up. As the videos progress, it seems that Jack is the more gifted twin, with Jill constantly trying to get his attention by hitting him, hurting girls around him, etc. The story shifts to the present, where an adult Jack (Adam Sandler) is a successful advertising executive in Los Angeles with a beautiful wife, Erin (Katie Holmes), and two kids, Sofie (Elodie Tougne) and Gary (Rohan Chand), while Jill (Sandler) never left the working-class neighborhood they grew up in. Jill recently inherited her childhood home, having lived with their mother until her death one year ago.

Glendon Wasey (Sean Penn) is a sleazy, down-on-his-luck con man struggling to sell glow-in-the-dark neckties in Shanghai. When he encounters the lovely Gloria Tatlock (Madonna), a missionary nurse who wants to obtain a supply of opium to ease the suffering of her patients, he decides to help her get hold of a stolen supply of the valuable drug. The only problem is that a lot of other people want to secure the stolen opium as well—gangsters, smugglers, thugs and a host of dangerous lowlifes.
The film is set in a post-apocalyptic future, where the vast majority of humanity has died due to a viral epidemic. The remaining humans have reverted to primitive tribalism.
A freak cyclone hits Los Angeles causing man-eating sharks to be scooped up in water spouts and flood the city with shark-infested seawater. Bar-owner and surfer Fin sets out with his friends, Baz, Nova, and George, to rescue his estranged wife, April, and their teenage daughter, Claudia, after the bar and boardwalk is destroyed in flooding. While heading to April's home, the group stops in a freeway to save people as flooding causes sharks to attack. George is killed and the group learns of a tornado warning. They arrive at April's house just before the first floor is flooded and shark-infested. Collin, April's boyfriend, is eaten by sharks, but the rest of the group escape unharmed.
Because of the repercussions left from his last adventure, Munchie must appear before a celestial court presided over by Kronus (Angus Scrimm). The tribunal proceeds to blame Munchie for a number of historical calamities, including the sinking of Atlantis, the crash of the Hindenburg, the cataclysm of Vesuvius, the meltdown of Chernobyl. As punishment, he is sentenced to help single mom Linda McClelland (Lesley-Anne Down). Linda's son Chris (Trenton Knight) is the only one who can see Munchie, and Munchie and he become best buddies. Chris is the pitcher of a Little League team, and is being bullied by Brett Carlisle (Corey Mendelsohn), who is his rival in baseball as well as for the attentions of the girl-next-door Jennifer (Natanya Ross). While out of town on a business trip, Linda's boss makes improper advances to her. When she refuses his advances, he fires her. This creates financial hardship on the family. Munchie intervenes in their lives, making it so Chris has a perfect game and impresses Jennifer, and Linda finds a bag of money to prevent eviction. Munchie appears back before the tribunal, who assign his next punishment... to help Bill Clinton.
A small-town manchild with big buck-teeth, Bucky Larson (Nick Swardson), stumbles upon a family secret: His quiet and reserved parents (Edward Herrmann and Miriam Flynn) were famous porn stars in the 1970s. This motivates him to leave northern Iowa for Hollywood, hoping to follow in their footsteps and fulfill his destiny as the biggest adult-film star in the world. Unfortunately, he has no idea how to become a porn star like his parents, and his penis is incredibly small.
There are three ongoing plots in the film. The primary one surrounds a white trash, trailer park family in which the daughter is sleeping with her mother's husband, prompting the mother to constantly try to outdo her promiscuous daughter's behavior out of spite, including sleeping with her daughter's boyfriend. The secondary plot revolves around an urban black woman whose boyfriend is sleeping with her two best friends, but the three are united against the boyfriend when he begins sleeping with the daughter of the above-mentioned family. The third plot revolves around Jerry and the show itself, detailing the difficulty Jerry faces in trying to come to terms with his rather dubious claim to fame, and the staff's utter amazement at the bizarre stories they must deal with.
