High-school senior Peter Parker visits a genetics laboratory with his friend Harry Osborn and love interest, Mary Jane Watson for a school field trip. There, Peter is bitten on the hand by a genetically engineered "super spider". Shortly after arriving home to his Aunt May and Uncle Ben, he goes unconscious. Meanwhile, Harry's father, scientist Norman Osborn, owner of Oscorp, is attempting to preserve his company's critically important military contract. He experiments on himself with a new but unstable performance-enhancing chemical vapor that increases his speed, strength, and stamina. However, it also drives him insane and he kills his assistant, Mendel Stromm. The next morning, Peter finds that his previously impaired vision has improved and his body has metamorphosized into a more muscular physique. At school, he finds that his body can produce webs and his quickened reflexes let him avoid injury during a confrontation with bully Flash Thompson. Peter discovers he has developed superhuman speed, strength, the ability to stick to surfaces, and a heightened ability to sense danger.

Sans que ses amis et compagnons d'aventures de l'épisode 2, Bethany, Fridge et Martha, ne soient au courant, Spencer a conservé les morceaux du jeu vidéo Jumanji dans le sous-sol de la maison de sa mère. De retour pour les vacances, alors qu'il mène une vie d'étudiant difficile à New-York et qu'il est contraint de partager sa chambre avec son grand-père convalescent, il répare le système et est happé par le jeu. Le lendemain, ses amis, avec qui il avait rendez-vous pour un brunch, passent chez lui prendre de ses nouvelles, ne l'y trouvent pas, mais constatent que le jeu en est en cours dans la cave de Spencer : tous décident de rentrer dans Jumanji pour le sauver. Le grand-père de Spencer, Eddie, et son ami Milo Walker entendent le chahut et se laissent entraîner par inadvertance dans le jeu et deviennent deux des avatars de Jumanji. Les amis adolescents doivent aider Eddie et Milo à s’habituer à leurs avatars du jeu, les aider à retrouver Spencer et à s'échapper à nouveau de Jumanji (ce qui n'est pas une tâche facile étant donné qu'il s'agit du niveau supérieur).

Kip Raines (Giovanni Ribisi), an aspiring car thief from Long Beach, is cruising with Mirror Man (T.J Cros) and Toby (William Lee Scott), looking for a Porsche 911 Carrera to steal. Former Army Ranger Cameron Poe is sentenced to prison for manslaughter for using excessive force on a drunk man while trying to protect his pregnant wife Tricia. Poe is paroled eight years later, and is to be released after being flown to Alabama on the Jailbird, a C-123K transport prison aircraft. Along with Poe are several other prisoners including his diabetic cellmate and friend Mike "Baby-O" O'Dell, who is being transferred (but not yet paroled) with Poe. The transfer is being overseen by U.S. Marshal Vince Larkin, as the transfer includes notorious criminal mastermind Cyrus "Cyrus The Virus" Grissom, gangster and Black Guerrilla member Nathan "Diamond Dog" Jones and mass murderer William "Billy Bedlam" Bedford for their transfer to a new Supermax prison. Larkin is approached at the last minute by DEA agents Duncan Malloy and Willie Sims, who ask for Sims to be brought aboard disguised as a prisoner so that he can extract more information from drug lord Francisco Cindino, a prisoner that is to be picked up at Carson City, Nevada en route. Larkin agrees, unaware that Malloy has hidden a gun on Sims' body.

In 1995, schemer Daniel Lugo (Mark Wahlberg) is a recently released convict who had served time for Medicare fraud. Sun Gym owner John Mese (Rob Corddry) hires him to increase membership and make the gym more fitness-based. Lugo increases the gym's membership by 75% within six weeks and befriends trainer Adrian Doorbal (Anthony Mackie), a bodybuilder who uses steroids, which render him impotent. Lugo soon lusts after the earnings and lifestyle achieved by Victor Kershaw (Tony Shalhoub), a sleazy, new gym member he begins to train who Lugo believes is a crook.
