The story is set in Edo (now Tōkyō) in Japan.
The film starts with a parade of samurai and their concubines (oiran, distinguished by their high shoes) along an avenue of cherry trees. Koide, called Seinosuke by his woman (Kotaro Bando), an artist/samurai apprenticed to a Kanō master, leaves the parade and visits a print shop where he sees a woodcut print by Utamaro that boasts of ukiyo-e 's superiority to the official style. Enraged, he goes to a tea-house to find Tsutaya Jūzaburō, the owner of the print shop, to express his displeasure. Word is leaked to Utamaro to avoid the tea-shop, but instead he goes there directly to investigate. Koide then challenges him to a duel. Utamaro counter-challenges him with a different kind of duel––a painting contest.

The film centers on Cisco Pike (played by Kris Kristofferson), an out of luck and out of work musician, and Sergeant Leo Holland (played by Gene Hackman). Cisco is a former drug dealer who has been busted several times by Sgt. Holland, and one night Holland comes to the door and demands that Cisco move a large amount of marijuana for him or suffer jail. He has several misadventures selling the marijuana, meets up with Doug Sahm at a recording studio, has his friend Jesse Dupree visit (played by Harry Dean Stanton, billed as H.D. When his ranch hands abandon him to join a gold rush, rancher Wil Andersen (John Wayne) is forced to find replacement drovers for his upcoming 400-mile (640 km) long cattle drive. He rides into deserted Bozeman, Montana. There, Anse Peterson (Slim Pickens) suggests using local schoolboys. Andersen visits the school but departs unconvinced. The next morning, a group of the boys show up at Andersen's ranch to volunteer for the drive. Andersen tests the boys' ability to stay on a bucking horse. As the boys successfully take turns, Cimarron (A Martinez), another young man slightly older than the others, rides up. After successfully subduing and riding the test horse, Cimarron gets into a fight with Slim (Robert Carradine), the oldest of the boys.
