On a rainy afternoon in Paris, debonair actor Phillipe Martin (Francis Lederer) goes to a darkened movie theatre for a romantic assignation with his married mistress, Yvonne (Liev De Maigret), but sits in the wrong seat and kisses instead lovely Monique Pelerin (Ida Lupino), the daughter of a powerful publisher (Joseph Cawthorn). Monique, who is engaged to powerful Count Alfredo Donstelli (Erik Rhodes), makes a public accusation against Phillip, and the priggish head of the Purity League (Eily Malyon) exploits the incident until it becomes a national scandal, with Phillipe dubbed "The Kissing Monster". When Phillipe is tried, his defense is that he was overcome by Monique's beauty, and that it is a Frenchman's nature to be romantic, even to perfect strangers. His punishment is to spend just three days in jail, but when he is released, he discovers that Monique has paid his fine, supposedly to avoid more publicity, but actually because she is secretly attracted to him.

Ranch owner Steve Stevens (John Van Pelt) is shot by his greedy partner, Martin (Lon Chaney, Jr.), who then sets fire to Stevens' barn. Stevens' last request is for cowboy Gene Autry (Gene Autry) to look after his daughter, Lou Ann (Ann Gillis), who was trampled by a horse during the barn fire and paralyzed. Martin befriends Lou Ann, hoping to gain possession of the secret gold mine beneath the Stevens' ranch. Lou Ann does not suspect Martin in her father's murder.

Wealthy murder mystery writer Paula Bradford (Jean Arthur) returns from her worldwide travels to see her former husband, surgeon Dr. Lawrence "Brad" Bradford (William Powell). He had divorced her because she was always involving him in real-life murder cases, but she wants him back. When a jockey riding the favorite dies while leading a race, she is convinced it was murder. She and Mike North (Frank M. Thomas), the horse's trainer, persuade the reluctant doctor to investigate. The musical's story spans about forty years, from the late 1880s to the late 1920s. Magnolia Hawks is an eighteen-year-old on her family's show boat, the Cotton Palace which travels the Mississippi River putting on shows. She meets Gaylord Ravenal, a charming gambler, falls in love with him, and eventually marries him. Together with their baby daughter, the couple leaves the boat and moves to Chicago, where they live off Gaylord's gambling winnings. After about ten years, he experiences an especially bad losing streak and leaves Magnolia, out of a sense of guilt that he is ruining her life because of his losses. Magnolia is forced to bring up her young daughter alone. In a parallel plot, Julie LaVerne (the show boat's leading actress, who is part African-American, but "passing" as white) is forced to leave the boat because of her background, taking Steve Baker (her white husband, to whom, under the state's law, she is illegally married) with her. Julie is eventually also abandoned by her husband, and she becomes an alcoholic. Magnolia's becomes a success on the stage in Chicago. Twenty-three years later Magnolia and Ravenal are reunited at the theater in which Kim, their daughter, is appearing in her first Broadway starring role.
